摘 要:旨在研究褪黑素对猪胎盘滋养层细胞(porcine placental trophoblast cells,pTr)增殖和功能的影响。不同浓度褪黑素(0、10 nmol/L、100 nmol/L、1μmol/L、10μmol/L、100μmol/L)处理pTr细胞48 h后,通过测定细胞数、VEGF、EGF、E2、P4分泌水平及相关基因mRNA表达水平,分析褪黑素对pTr细胞增殖及功能的影响。结果显示,处理48 h时,10μmol/L的褪黑素显著促进pTr增殖(P<0.05),并显著提高VEGF、EGF、E2和P4分泌水平(P<0.05)。与空白对照组相比,10μmol/L褪黑素显著上调MT1、CDK4、SLC2A1、VEGF及EGFR的表达(P<0.05),并极显著上调VEGFR2、EGF、ESR1和PGR的表达(P<0.01)。综上所述,10μmol/L的褪黑素能显著促进pTr增殖,其可能通过与MT1结合直接,或通过上调CDK4、SLC2A1基因表达间接促进pTr细胞的增殖;此外,褪黑素还可能通过上调pTr中生长因子及其受体、类固醇激素受体等基因表达,以及促进生长因子、生殖激素合成和分泌改善猪胎盘功能。To investigate the effects of melatonin on the proliferation and function of porcine placenta trophoblast cells(pTr),pTr cells were treated with different concentrations of melatonin(0,10 nmol/L,100 nmol/L,1μmol/L,10μmol/L and 100μmol/L)for 48 hours.The effect of melatonin on the proliferation and function of pTr cells was analyzed by detecting the number of cells,the secretion levels of VEGF,EGF,E2,P4 and the mRNA expression levels of related genes.The results showed that the concentration of 10μmol/L melatonin significantly promoted the proliferation of pTr(P<0.05).The secretion levels of VEGF,EGF,E2 and P4 were significantly increased(P<0.05).Compared with the blank control group,10μmol/L melatonin significantly up-regulated the expression of MT1,CDK4,SLC2A1,VEGF,and EGFR in pTr cells(P<0.05),and the expression of VEGFR2,EGF,ESR1,and PGR(P<0.01).In conclusion,10μmol/L melatonin could promote the proliferation of pTr cells by directly binding to MT1,or indirectly by regulating the expression of CDK4 and SLC2A1 genes.In addition,melatonin might also improve placental function in pigs by regulating the expression of growth factors and their receptors and steroid hormone receptors and promoting the synthesis and secretion of growth factors and reproductive hormones.
