摘 要:为探讨作为传统溴系阻燃剂十溴二苯醚(BDE-209)替代品的新型阻燃剂十溴二苯乙烷(DBDPE)暴露对小鼠肝肾功能的影响,将60只雄性小鼠随机分为5组,每天分别给予低、中、高剂量(5、50、500 mg/kg体重)的DBDPE,对照组给予相同体积的玉米油,联合染毒组每天给予50 mg/kg体重的DBDPE和BDE-209,每天灌胃1次,连续暴露60 d。试验期间观察小鼠的一般体征,测量小鼠体质量,末次染毒后测定小鼠血清生化指标、肝脏和肾脏系数,通过苏木素-伊红(HE)、油红O和刚果红染色观察小鼠肝脏和肾脏组织病理学变化。结果显示,与对照组相比,染毒组小鼠临床体征未见异常,体质量无显著变化;高剂量组小鼠肝脏系数显著升高(P<0.05),中剂量组小鼠肾脏系数极显著降低(P<0.01)。高剂量组小鼠血清中尿素(UA)含量极显著升高(P<0.01),中剂量组小鼠血清中白蛋白(ALB)含量和碱性磷酸酶(ALP)活性极显著升高(P<0.01),甘油三酯(TG)含量降低(P<0.05)。组织病理学结果可见,各攻毒组小鼠肝脏细胞均发生不同程度的脂肪变性和坏死,高剂量组小鼠肾脏间质有大量淀粉样物质沉着。结果表明,每天500 mg/kg体重DBDPE暴露导致小鼠肝肾组织结构改变,诱导小鼠肝肾组织细胞代谢紊乱和功能障碍,对肝肾具有毒性效应。To investigate the effects of decabromodiphenyl ethane(DBDPE),a novel flame retardant which instead of the traditional brominated flame retardants(BDE-209),on liver and kidney functions of mice.In this experiment,60 male mice were randomly divided into five groups,the combined group was given 50 mg/kg bw/day dose of DBDPE and BDE-209,low,medium and high dose groups were given 5,50 and 500 mg/kg bw/day of DBDPE,respectively.the control group was given same volume corn oil.The exposure was administered once a day for 60 consecutive days.Throughout the experiment,the general signs of mice were observed,body weight was measured,and following the final exposure,serum biochemical indicators,liver and kidney indexes were determined.Histopathological changes in the liver and kidney tissues of the mice were observed through Hematoxylin and Eosin(HE),Oil Red O,and Congo Red staining.The results showed that compared with the control group,the clinical signs in the treated mice showed no abnormalities,and there were no significant changes in body weight.However,in the high-dose group,the liver index of mice significantly increased(P<0.05),the kidney index of mice in medium dose group mice exhibited extremely significant decrease(P<0.01).Moreover,the urea(UA)content in the serum of high-dose group mice was extremely significantly increased(P<0.01),and the albumin(ALB)content and alkaline phosphatase(ALP)activity in the serum of medium-dose group mice were extremely significant increased(P<0.01),and the triglyceride(TG)content was significantly decreased(P<0.05).The histopathological results showed that the hepatoceytes in each challenge group mice exhibited varying degrees of steatosis and necrosis,while the kidney tissues of the high-dose group mice exhibited a large amount of amyloid-like substance deposition.The results showed that exposure to 500 mg/kg bw/day DBDPE resulted in structural changes in the liver and kidney tissues of mice,causing metabolic disorders and dysfunction of liver and kidney,thus indicating a toxic
