摘  要:本研究以黄叶和绿叶‘鑫叶栾’(Koelreuteria paniculata‘Xinye’)半同胞子代为材料,测定了它们在7月和10月的色差、光合作用指数、叶绿素荧光参数等生理指标。此外还进行了转录组测序及RT-qPCR试验。结果表明,在测试的两个月份中,绿叶子代的色素含量均高出黄叶苗近2倍。此外,除绿叶子代的胞间CO_(2)浓度和光系统Ⅱ(PSⅡ)最大光化学效率低于黄叶子代外,其余各指标均高于黄叶子代。与7月份的材料相比,10月份黄叶和绿叶子代的色素、淀粉含量、光合能力均降低,可溶性糖含量和PSⅡ最大光化学效率升高。转录组分析表明,黄叶苗呈色原因可能与叶绿素合成降低、叶绿体发育不良及类黄酮合成等通路的相关基因表达量下调有关。此外,RT-qPCR结果表明了转录组数据的可靠性。本研究为进一步探究植物叶色形成的机理和创制彩叶植物新种质提供了新的见解。The half-sib progenie Koelreuteria paniculata‘Xinye’with yellow and green leaves were used as material.The physiological indexes of the plant materials sampled in July and October were measured,including color variation,photosynthesis indicators,chlorophyll fluorescence parameters,etc.Moreover,RNA-seq and RT-qPCR assays were also performed.The results show that the pigment contents of all green-leaf seedlings were nearly twice as high as those of yellow-leaf seedlings.Additionally,only two indexes(i.e.,C_(i) and F_(v)/F_(m))were lower in green-leaf seedlings than in the yellow-leaf seedlings,while all other physiological indexes were higher in green-leaf seedlings.Furthermore,compared to the plant materials sampled in July,the pigment and starch contents and photosynthesis ability were down-regulated,whereas the soluble sugar content and F_(v)/F_(m) were up-regulated in the plant materials sampled in October.Tran-scriptome results suggest that the yellow leaves of the seedlings may be attributed to the down-regulation of genes associated with pathways such as chlorophyll biosynthesis,chloroplast development,and flavonoid biosynthesis.RT-qPCR assay further confirmed the reliability of the RNA-seq data.This work provides insight into further investigating the mechanisms of the leaf color formation of the plants as well as creating new germplasm with colorful foliage.
