摘  要:以蒙古冰草×航道冰草种间杂种F1代幼穗为外植体,对影响愈伤组织诱导、分化和生根的激素浓度进行筛选,并建立了完整植株的再生体系。结果表明,杂种F1代的最适愈伤组织诱导培养基为MS+2,4-二氯苯氧乙酸(2,4-D)1.5 mg/L+6-苄氨基嘌呤(6-BA)0.3 mg/L,出愈率为81.7%;愈伤组织分化的培养基为6-BA 2.5 mg/L+萘乙酸(NAA)0.4 mg/L,分化率为71.7%;生根培养基为1/2 MS+NAA 0.2 mg/L,生根率为88.3%。经炼苗后,各基质移栽成活率大于90%。本研究通过建立蒙古冰草×航道冰草杂种F1代组织培养再生体系,为有效保存冰草杂种F1代新种质,并为后期秋水仙碱诱导其染色体加倍恢复育性奠定基础。The F 1 young panicle of the interspecific hybrid of Agropyron mongolicum Keng×Agropyron cristatum cv.Fariway was selected as explants to screen the hormone concentration affecting callus induction,differentiation and rooting,and a complete plant regeneration system was established.The results showed that the optimal callus induction medium of hybrid F 1 generation was MS+2,4-dichlorophenoxyacetic acid(2,4-D)1.5 mg/L+6-benzylaminopurine(6-BA)0.3 mg/L,and the healing rate was 81.7%.The medium for callus differentiation was 6-BA 2.5 mg/L+naphthalene acetic acid(NAA)0.4 mg/L,and the differentiation rate was 71.7%.The rooting medium was 1/2 MS+NAA 0.2 mg/L,and the rooting rate was 88.3%.After refining the seedlings,the survival rate of each substrate transplanting was more than 90%.In this study,the F 1 generation tissue culture regeneration system of Agropyron mongolicum Keng×Agropyron cristatum cv.Fariway hybrid was established to effectively preserve the F 1 generation and laid a foundation for the recovery of chromosome doubling induced by colchicine in the later stage.
