摘　　要：针对目前多种生物类分析仪器羧基荧光素(FAM)通道荧光校准缺乏合适的荧光标准溶液的问题,制备了5种浓度的FAM荧光标准溶液,并对其制备方法、荧光性能、稳定性、均匀性等方面进行了考察。结果表明,该荧光标准溶液荧光性能优于常规FAM溶液,均匀性良好,在25、4、-20℃3个温度下均可稳定保存7个月。通过对恒温扩增核酸分析仪、数字PCR仪、多功能酶标仪等多种仪器进行校准,证明其具有良好的适用性,为FAM通道的荧光校准提供了可靠的参考依据。In view of the lack of suitable fluorescent standard solutions for fluorescence calibration of carboxyfluorescein(FAM)channels in various bioanalytical instruments,five standard solutions of FAM fluorescence with different concentrations were prepared,and their preparation method,fluorescence performance,stability,uniformity and other aspects were investigated.The results showed that the fluorescence performance of the standard solutions was better than that of conventional FAM solution,and can be stably stored at 25,4,and-20℃for 7 months with maintaining good uniformity.Through the calibration of various instruments such as isothermal amplification nucleic acid analyzers,digital PCR machines,and multimode reader,the good applicability of the fluorescent standard solutions has been demonstrated,providing reliable reference for fluorescence calibration in FAM channel.
