摘 要:为了检验自拟中药组方“益宫清”的抑菌和抗炎效果,并将其制成新型中药泡腾栓剂,试验采用试管二倍稀释法检测中药“益宫清”对子宫内膜炎常见致病菌的最低抑菌浓度(MIC)和最低杀菌浓度(MBC);选择雌性C57小鼠32只,随机分为对照组、大肠杆菌组、“益宫清”中药组和恩诺沙星组,每组8只,对照组子宫灌注生理盐水,其余组灌注大肠杆菌建立子宫内膜炎模型,每24 h 1次,连续3次。首次造模24 h后,“益宫清”组灌胃中药液,恩诺沙星组灌胃恩诺沙星,每24 h 1次,连续2次。最后1次治疗24 h后处死小鼠,检测小鼠子宫指数;采用H.E.染色观察子宫组织病理变化;采用ELISA法检测子宫组织白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)等相关炎症因子质量浓度。筛选合适的栓剂基质,利用正交试验筛选最佳基质配比,制成栓剂并进行质量检查。结果表明:“益宫清”水提取物对子宫内膜炎常见致病菌有一定的抑制作用,其中对无乳链球菌的抑制效果最强,MIC为3.91 mg/mL,MBC为7.81 mg/mL,然后依次是金黄色葡萄球菌、大肠杆菌。与对照组相比,大肠杆菌组子宫指数极显著增加(P<0.01);与大肠杆菌组相比,“益宫清”组子宫指数极显著降低(P<0.01),恩诺沙星组差异不显著(P>0.05);且“益宫清”水提取物能够缓解由大肠杆菌导致的子宫组织病理性损伤。与对照组相比,大肠杆菌组子宫组织中TNF-α、IL-1β质量浓度极显著升高(P<0.01),IL-6质量浓度显著升高(P<0.05);与大肠杆菌组相比,“益宫清”组子宫组织中TNF-α、IL-1β、IL-6质量浓度极显著降低(P<0.01),恩诺沙星组子宫组织中TNF-α、IL-6质量浓度显著降低(P<0.05)。泡腾栓优选配比为PEG-60006.8 g,柠檬酸1.2 g,碳酸氢钠1.6 g,主药含量2.4 g,质量检查合格。说明“益宫清”具有一定的抑菌和抗炎效果,并成功将其制成泡腾栓剂。In order to test the antibacterial and anti-inflammatory effects of the self-developed traditional Chinese medicine formula“Yigongqing”,which could be made into a new type of Chinese medicine effervescent suppository,in the test,determination of the minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)of“Yigongqing”against common endometritis-causing pathogens was carried out by tube double dilution method.Thirty-two female C57 mice were randomly divided into four groups,namely control group,E.coli group,“Yigongqing”group,and enrofloxacin group,with eight mice in each group.In the control group,the uterus was filled with saline;the rest of the groups was filled with E.coli to establish the endometritis model every 24 h for three consecutive times.Twenty-four hours after the first modeling,the“Yigongqing”group was gavaged with Chinese herbal medicine;the enrofloxacin group was gavaged with enrofloxacin every 24 h for two consecutive times.The mice were executed 24 h after the last treatment,and the uterine index was detected;H.E.staining was used to observe the pathological changes of uterine tissues.Uterine tissue mass concentrations of interleukin-1β(IL-1β),interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)and other related inflammatory factors were detected by ELISA.A suitable suppository matrix was screened;the optimal ratio of the matrix was screened by orthogonal test;the suppository was made and quality was checked.The results showed that the aqueous extract of the“Yigongqing”had a certain inhibitory effect on the common pathogenic bacteria of endometritis;among them,the strongest inhibitory effect was on Streptococcus agalactiae,with the MIC of 3.91 mg/mL and MBC of 7.81 mg/mL,then followed by Staphylococcus aureus,Escherichia coli.The uterine index was highly significantly increased in the E.coli group compared to the control group(P<0.01).Compared with the E.coli group,the uterine index of the“Yigongqing”group was significantly reduced(P<0.01);the
