从患病的大口黑鲈检出疱疹病毒

摘　　要：采用超微病理学电镜观察和PCR检测等方法,首次从患病的养殖大口黑鲈(Micropterus salmoides)检出疱疹病毒。病鱼组织超薄切片透射电镜观察结果显示,鳃组织细胞内存在直径约70 nm-80 nm的圆形或双环状核衣壳,空心或小部分核心致密;肝组织细胞内质网所查见部分核衣壳有纤突。所检出病毒的形态学和形态发生学特征符合疱疹病毒目(Order Herpesvirales)的病毒形态。采用鳗鲡疱疹病毒的特异性引物对病毒进行PCR方法检测,从24份患病大口黑鲈的鳃、肝、脾、肾组织混样中检出13份阳性结果。基于大口黑鲈疱疹病毒毒株DNA聚合酶基因片段序列的系统进化树分析,进一步证明该病毒可能是鳗鲡疱疹病毒的成员。Herpesvirus was first detected from diseased cultured largemouth bass(Micropterus salmoides)by the ultrastructual pathological study and PCR detection.Herpesvirus was detected from the cell of gill and liver of the diseased Largemouth bass by electron microscope examination.Virions in gill epithelial cells,intestinal epithelial and liver cells were round,double-shell in shape or hollow and about 70 nm to 80 nm in diameter.Around the nucleus of the liver cell there were envelopes with diameter about 100 nm.The morphological and morphogenetic characteristics of the virus detected from the cultured Largemouth bass are comparable with the morphology of the family of Herpesviridae.We investigated in 9 farms,24 samples of diseased cultured Largemouth bass were detected by PCR,13 samples were positive for herpesvirus.Based on the sequence of DNA polymerase gene fragment of the herpesvirus strain of largemouth bass,the phylogenetic tree analysis further proves that the virus may be a member of the genus Cyprinus herpesvirus.
