摘  要:建立基于高分辨率融解曲线(High resolution melting,HRM)的快速检测新冠病毒变异株的方法,为新冠变异株快速检测提供技术支撑。针对新冠病毒S基因的氨基酸突变位点G339H/D、D796Y、V83A设计HRM引物,将新冠病毒实时定量荧光PCR(RT-PCR)阳性的核酸逆转录后进行HRM分析,并对33份新冠病毒RT-PCR阳性核酸标本进行检测,与二代测序结果进行比较。建立的HRM方法能区分Delta、Omicron及XBB亚型,33份标本的HRM分析结果显示其中2份为Delta株,31份为Omicron株,12份为XBB亚型。HRM分析结果与二代测序结果一致。本实验建立的HRM方法能够准确检测出新冠病毒变异株中的Delta、Omicron及XBB亚型,可应用于快速鉴定新冠病毒型别。To:(i)establish a method based on high-resolution melting(HRM)curves for the rapid detection of severe acute respiratory-coronavirus-2(SARS-CoV-2)variants;(ii)provide technical support for the rapid detection of SARS-CoV-2 variants.HRM primers were designed for the amino-acid mutation sites G339H/D,D796Y,and V83A of the S gene of SARS-CoV-2.Detection of the nucleic acids of SARS-CoV-2 was undertaken with real-time reverse transcription-quantitative polymerase chain reaction.Thirty-three samples of SARS-CoV-2-positive nucleic acids were detected and compared with the results of next-generation sequencing.The HRM method could be used to distinguish the Delta,Omicron,and XBB variants of SARS-CoV-2.HRM analyses of 33 samples showed that there were two Delta,31 Omicron,and 12 XBB subtypes of SARS-CoV-2.The results of HRM analyses were consistent with those of next-generation sequencing.Our HRM method could be used to accurately detect the Delta,Omicron,and XBB variants of SARS-CoV-2.
