健脾益智法调控CXCL1/CXCR2通路介导中性粒细胞局部浸润促进MCAO大鼠轴突再生机制研究

摘  要:目的:研究健脾益智法对MCAO大鼠大脑皮质层和海马区中性粒细胞浸润情况、脑组织中CXCL1、CXCR2表达水平及轴突再生情况,探讨脾脑相关理论的微观机制。方法:将20只SD大鼠按照随机数字表法分为正常组、模型组、中药组,除正常组外,其余两组参照国外学者Zea Longa的线栓法建立MCAO大鼠模型,造模成功后中药组给予健脾益智汤连续灌胃,各组分别于术后7 d、14 d采用醋酸AS-D萘酚酯酶组织化学染色检测大鼠大脑皮质及海马中性粒细胞浸润情况,免疫荧光进行脑组织中CXCL1、CXCR2表达水平及轴突再生情况检测。结果:中药组术后7d、术后14d大鼠大脑皮质层和海马区中性粒细胞数量与同期模型组、正常组比较显著减少,中药组术后7 d、14 d CXCL1和CXCR2表达量均较模型组明显增高。与模型组比较,中药组术后7 d、14 d轴突再生明显,术后14 d尤甚。结论:健脾益智法可能激活CXCL1/CXCR2通路,促进轴突再生,从而修复受损的神经。Objective To study the neutrophil infiltration in the cerebral cortex,CXCL1and CXCR2 expression levels and axonal regeneration in MCAO rats,and to investigate the microscopic mechanism of spleen and brain.Methods 20 SD rats are divided into normal group,model group,Chinese medicine group,except normal group,the rest two groups by foreign scholars Zea Longa MCAO rat model,mold after successful traditional Chinese medicine group give spleen puzzle soup continuous gastric,each group in 7 days,14 days respectively using acetate AS-D naphthol esterase histochemical staining to detect the infiltration of rat cerebral cortex and hippocampus,immunofluorescence for CXCL1,CXCR 2 expression level and axonal regeneration.Results The number of neutrophils in the cerebral cortex and hippocampus of 7 and 14 days were significantly decreased compared with the concurrent model group;CXCL 1 and CXCR 2 at 7 and 14 days were significantly higher compared with the model group.Compared with the model group,axon regeneration was obvious in 7 days and 14 days after surgery,especially in 14 days after surgery.Conclusion The method may activate CXCL1/CXCR 2 pathway and promote axon regeneration to repair the damaged nerves.
