摘  要:目的:建立鹅不食草配方颗粒指纹图谱检测方法及一测多评含量测定。方法:采用Agilent Poroshell 120 SB-Aq色谱柱(2.1 mm×150 mm,1.9μm),乙腈-0.1%磷酸为流动相,梯度洗脱,流速0.2 mL/min;柱温为25℃;0~60 min检测波长为325 nm,60~100 min检测波长为225 nm。结果:指纹图谱确定26个共有峰,多批次图谱相似度均大于0.9。建立含量测定方法线性范围良好(r^(2)=0.9994~1.000),平均加样回收率99.6%~101.1%建立相对校正因子,通过绿原酸能够准确计算含量,咖啡酸、3,5-二-O-咖啡酰奎宁酸、4,5-二-O-咖啡酰奎宁酸、3,4-二-O-咖啡酰奎宁酸、隐绿原酸、新绿原酸相对校正因子为1.761、1.387、1.273、0.828、1.141、1.082。结论:本研究制定的方法能作为检测手段简便有效、重复效果好,可为鹅不食草配方颗粒的质量控制和评价提供依据。Objective To establish a fingerprint detection method of Centipeda Minima Dispensing Granules(CMDG)formula particles and a multi-evaluation content determination.Methods The chromatography was performed on Agilent Poroshell 120 SB-Aq column(2.1 mm×150 mm,1.9μm)with acetonitrile-0.1%phosphoric acid as mobile phase by gradient elution at the flow rate of 0.2mL/min.The column temperature was 25℃.The detection wavelength of 0-60 minutes is 325nm,and the detection wavelength of 60-100 minutes is 225nm.Results The 26 common peaks were identified by fingerprint,and the similarity of multiple batches was greater than 0.9.The linear range of the method was good(r^(2)=0.9994-1.000),and the average recovery rate was 99.6%-101.1%.The relative correction factors of caffeic acid,3,5-O-Dicaffeoylquinic Acid,4,5-O-Dicaffeoylquinic Acid,3,4-O-Dicaffeoylquinic Acid,Cryptochlorogenic acid and Neochlorogenic acid were 1.761,1.387,1.273,0.828,1.141,1.082.Conclusion The method developed in this study can be used as a simple and effective detection method with good repeatability,and can provide a basis for the quality control and evaluation of CMDG.
