摘 要:为研究双组分系统bfmR基因对鲍曼不动杆菌生物学特性的影响,利用RecA同源重组技术构建鲍曼不动杆菌临床分离株AB43双组分系统bfmR基因缺失突变株并观察其生物学性状的变化,采用ExPASY-ProtParam、SMART、SOPMA和SWISS-MODEL等软件对bfmR进行生物信息学分析以及系统进化树构建。结果表明:与鲍曼不动杆菌野生株AB43相比,bfmR基因缺失突变株(AB43ΔbfmR)生长速度明显降低,细菌的自聚合能力和胞内超氧化物歧化酶(SOD)活性明显下降,生物被膜形成能力、黏附和入侵能力无显著性变化,而对头孢曲松的敏感性明显提高。生物信息学分析结果显示,bfmR蛋白为较稳定的亲水性蛋白,二级结构中α-螺旋含量最多,占总数的36.13%;鲍曼不动杆菌AB43株bfmR基因与标准株ATCC 17978和ATCC 19606的亲缘关系较近。综上,鲍曼不动杆菌AB43ΔbfmR的构建为进一步探讨双组分系统bfmR基因的调控机制提供试验基础。To study the effect of two-component system bfmR gene on the biological characteristics of Acinetobacter baumannii,the bfmR gene deletion mutant of A.baumannii strain AB43 was constructed using RecA homologous recombination and the changes of biological characteristics were observed.The bfmR gene was analyzed by ExPASY-ProtParam,SMART,SOPMA and SWISS-MODEL.The results showed that the growth rate of bfmR gene deletion mutant(AB43ΔbfmR)was significantly reduced,the autopolymerization ability and intracellular superoxide dismutase(SOD)activity of the bacteria were significantly decreased,the biofilm forming ability,adhesion and invasion ability were not significantly changed,and the sensitivity to ceftriaxone was significantly increased.The bioinformatics analysis results showed that the bfmR protein was a relatively stable hydrophilic protein,alpha-helix was the highest in the secondary structure,accounting for 36.13%of the total.The bfmR gene of A.baumannii strain AB43 was closely related to that of the standard strain ATCC 17978 and ATCC 19606.In conclusion,the construction of A.baumannii AB43ΔbfmR provides an experimental basis for further exploration of the regulatory mechanism of bfmR gene in the two-component system.
