青藤碱对骨关节炎软骨细胞增殖和炎症的影响

摘  要:目的:明确青藤碱对OA软骨细胞增殖和炎症的影响并探究其作用机制。方法:采用10ng/mL白介素1β(IL-1β)诱导软骨细胞建立骨关节炎软骨细胞损伤模型,加入不同浓度的青藤碱处理细胞;采用CCK-8检测细胞增殖能力;采用ELISA检测IL-6、TNF-α的水平;采用Western blot检测IL-6、TNF-α、Wnt和β-catenin蛋白的表达量;采用qRT-PCR检测IL-6、TNF-α、Wnt和β-catenin表达量。结果:不同浓度青藤碱均可逆转IL-1β诱导导致的软骨细胞增殖活性下降(P<0.05),但组间比较无明显差异(P>0.05);不同浓度青藤碱还可下调IL-6、TNF-α炎症因子水平,低、高浓度青藤碱下调更显著(P<0.05),但低、高浓度组间比较无明显差异(P>0.05);低浓度青藤碱可降低软骨细胞中IL-6、TNF-α的蛋白和基因表达水平,还可下调Wnt和β-catenin蛋白和基因表达,差异有统计学意义(P<0.05);但中、高浓度青藤碱无明显降低IL-6、TNF-α、Wnt和β-catenin的蛋白和基因表达,差异无统计学意义(P>0.05)。结论:低浓度青藤碱可促进软骨细胞增殖并抑制OA软骨细胞的炎症,部分可能与调控Wnt/β-catenin信号通路有关。Objective:To investigate the effect of Sinomenine on the proliferation and inflammation of OA chondrocytes,and to explore its specificmechanism.Methods:Chondrocytes were induced by 10ng/mL interleukin 1β(IL-1β)to establish a model of osteoarthritis chondrocyte injury,anddifferent concentrations of sinomenine were added to treat the cells.CCK-8 assay was used to detect cell proliferation;ELISA was used to detect thelevels of IL-6 and TNF-α;Western blot was used to detect the expression of IL-6,TNF-α,Wnt andβ-catenin proteins;The expressions of IL-6,TNF-α,Wnt andβ-catenin were detected by qRT-PCR.Results:Different concentrations of sinomenine could reverse the decrease of chondrocyte proliferationactivity induced by IL-1β,and the difference was statistically significant(P<0.05),but there was no significant difference between the groups(P>0.05).At the same time,the levels of IL-6 and TNF-αinflammatory factors were also downregulated by different concentrations of sinomenine,and the downregulation of low-concentration and high-concentration sinomenine was more significant,and the difference was statistically significant(P<0.05),but there was no significant difference between the low-concentration and high-concentration groups(P>0.05).Low concentration of sinomeninecould reduce the protein and gene expression levels of IL-6 and TNF-αin chondrocytes,and also down-regulate the protein and gene expressionof Wnt andβ-catenin,and the difference was statistically significant(P<0.05).However,the protein and gene expressions of IL-6,TNF-α,Wntandβ-catenin were not significantly reduced at medium and high concentrations,and the difference was not statistically significant(P>0.05).Conclusion:Low concentrations of sinomenine may promote chondrocyte proliferation and inhibit inflammation in OA chondrocytes,which may be partlyrelated to the regulation of the Wnt/β-catenin signalling pathway.
