摘  要:目的:探究枸杞多糖(LBP)和亚硒酸钠(Na_(2)SeO_(3))对镉(Cd)致大鼠肝毒性的拮抗作用。方法:40只SD大鼠随机分为对照组、CdCl_(2)组、LBP组、Na_(2)SeO_(3)组、LBP联合Na_(2)SeO_(3)组,每组8只。对照组不造模,其余四组腹腔注射CdCl_(2)染毒。染毒1 h后,对照组和CdCl_(2)组给予生理盐水灌胃,LBP组、Na_(2)SeO_(3)组分别给予LBP、Na_(2)SeO_(3)灌胃,LBP联合Na_(2)SeO_(3)组给予LBP和Na_(2)SeO_(3)灌胃,每日染毒和灌胃各进行1次,共干预35 d。观察各组大鼠肝脏病理改变,测定大鼠肝组织Cd含量;检测血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、直接胆红素(DB)及总胆红素(TB)的含量以及肝组织超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和丙二醛(MDA)含量;Western blot检测肝组织中苹果酸脱氢酶(MDH)、琥珀酸脱氢酶(SDH)、铁调节蛋白(IRP-1)和谷胱甘肽过氧化物酶4(GPX4)的蛋白表达水平。结果:与CdCl_(2)组、LBP组和Na_(2)SeO_(3)组相比,LBP联合Na_(2)SeO_(3)组肝细胞排列更紧密,细胞核更清晰可见,肝小叶中央静脉无淤血。相比CdCl_(2)组,LBP联合Na_(2)SeO_(3)组能降低Cd在大鼠肝脏的蓄积(P<0.05)。LBP联合Na_(2)SeO_(3)组与CdCl_(2)组相比,血清ALT、AST水平降低(P<0.05);LBP联合Na_(2)SeO_(3)组与CdCl_(2)组相比,大鼠肝脏组织中SOD、GSH-Px水平显著升高,MDA降低(P<0.05);LBP联合Na_(2)SeO_(3)组大鼠肝脏组织中MDH、SDH、IRP-1、GPX4的蛋白表达水平较CdCl_(2)组、LBP组和Na_(2)SeO_(3)组升高(P<0.05)。结论:LBP和Na_(2)SeO_(3)联合干预可能通过增强大鼠肝脏的抗氧化能力拮抗Cd诱导的肝损伤。Objective:To explore the antagonistic effect of Lycium barbarum polysaccharide(LBP)and sodium selenite(Na_(2)SeO_(3))on hepatotoxicity induced by cadmium(Cd)in rats.Methods:Forty SD rats were randomly divided into control group,CdCl_(2) group,LBP group,Na_(2)SeO_(3) group and LBP combined Na_(2)SeO_(3) group,with eight rats in each group.The control group was not molded,and the other four groups were infected by intraperitoneal injection of CdCl_(2).1 h after exposure,the control group and CdCl_(2) group were given normal saline intragastric administration.LBP group and Na_(2)SeO_(3) group were given LBP and Na_(2)SeO_(3) by gavage.LBP combined with Na_(2)SeO_(3) group was given LBP and Na_(2)SeO_(3) by gavage.The patients were treated once a day with poison and once a day with intragastric administration for 35 days.The liver pathological changes of each group were observed and the content of Cd in liver tissue was determined.The contents of alanine aminotransferase(ALT),aspartate aminotransferase(AST),direct bilirubin(DB)and total bilirubin(TB)in serum and the contents of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and malondialdehyde(MDA)in liver tissue were detected.Western blot was used to detect the protein expression levels of malate dehydrogenase(MDH),succinate dehydrogenase(SDH),iron regulatory protein(IRP-1),and glutathione peroxidase 4(GPX4)in liver tissue.Results:Compared with CdCl_(2) group,LBP group and Na_(2)SeO_(3) group,the liver cells in LBP combined with Na_(2)SeO_(3) group were more closely arranged,the nucleus was more clearly visible,and the central vein of the hepatic lobule was not stasis;compared with CdCl_(2) group,LBP combined with Na_(2)SeO_(3) group could reduce the accumulation of Cd in the liver of rats(P<0.05);serum ALT and AST levels in LBP combined with Na_(2)SeO_(3) group were lower than those in CdCl_(2) group(P<0.05);compared with CdCl_(2) group,the levels of SOD and GSH-Px in liver tissues of LBP combined with Na_(2)SeO_(3) group were significantly increased,and M
