摘 要:目的:研究药物替诺福韦(TDF)对人肝星状细胞(LX-2)的CC趋化因子受体2(CCR2)表达的影响,探讨TDF在肝纤维化疾病发展中发挥抗纤维化的作用机制。方法:以LX-2作为体外实验研究对象,使用HE染色观察用药后细胞增殖情况。利用蛋白质印迹法检测使用不同浓度TDF和CCR2小干扰RNA(siRNA)对LX-2细胞α平滑肌肌动蛋白(α-SMA)及CCR2蛋白表达水平的影响。结果:siRNA沉默CCR2表达可显著下调肝纤维化相关蛋白α-SMA表达(P<0.01);TDF抑制LX-2细胞的增殖活化,明显降低细胞CCR2及α-SMA的蛋白表达水平(P<0.01)。结论:TDF可通过抑制CCR2表达抑制肝星状细胞活化,发挥抗肝纤维化作用。Objective To investigate the effect of TDF on the expression of CCR2 in human hepatic stellate cells LX-2,and explore the mechanism by which TDF plays an anti fibrotic role in the development of liver fibrosis diseases.Methods Human hepatic stellate cell LX-2 was used as the experimental research object in vitro.Observed the cell proliferation after medication using HE cell staining.Used Western blot method to detect the effects of different concentrations of TDF and siRNA on the expression levels ofα-SMA and CCR2 proteins in LX-2 cells.Results Silencing CCR2 by siRNA significantly downregulated the expression level of liver fibrosis related proteinα-SMA(P<0.01).TDF inhibited the proliferation and activation of LX-2 cells,significantly reduced the expression levels of CCR2 andα-SMA in cells(P<0.01).Conclusions TDF can inhibit the activation of hepatic stellate cells by downregulating the expression of CCR2,exerting an anti fibrosis effect.
