摘 要:目的构建广陈皮标准汤剂HPLC指纹图谱,并考察其与广陈皮药材、饮片的质量相关性。方法通过高效液相色谱法对广陈皮药材、饮片及标准汤剂各15批进行分析,采用中药色谱指纹图谱相似度评价系统(2012版)进行其指纹图谱的构建及相似度分析,并对三者的4种有效成分橙皮苷、川陈皮素、橘皮素和D-柠檬烯进行定量测定。结果建立了广陈皮药材、饮片及标准汤剂的HPLC指纹图谱,分别确定了18、18和16个共有峰。与广陈皮药材对照指纹图谱比较,饮片对照指纹图谱的相似度为0.994,标准汤剂对照指纹图谱的相似度为0.833。广陈皮标准汤剂的出膏率均值为(43.2±0.1)%,橙皮苷、川陈皮素、橘皮素和D-柠檬烯的转移率平均值分别为(19.3±4.9)%、(25.3±4.0)%、(16.6±2.5)%、(2.2±2.2)%。结论建立的指纹图谱反映出广陈皮药材、饮片及标准汤剂的化学成分基本一致。经分析比较,标准汤剂中橙皮苷、川陈皮素和橘皮素等成分与药材、饮片具有良好的正相关性。本研究可为广陈皮标准汤剂及配方颗粒等相关制剂的生产制备、质量控制提供参考。Objective To establish an HPLC fingerprints of Guangchenpi(dried tangerine peel)standard decoction and determine the quality correlation with its herbs and decoction pieces.Methods Totally 15 batches of Guangchenpi herbs,decoction pieces and standard decoction were analyzed by high performance liquid chromatography(HPLC).Their fingerprints were established and analyzed for similarity with Chinese Medicine Chromatographic Fingerprint Similarity Evaluation System(2012 version).The four active ingredients(hesperidin,nobiletin,tangeretin and D-limonene)were quantified.Results HPLC fingerprints of Guangchenpi,its decoction pieces and its standard decoction were obtained,and 18,18 and 16 common peaks were identified.The similarity of decoction pieces was 0.994 when compared with the control fingerprint profiles of its herbs,while the similarity of standard decoction was 0.833.In addition,the mean value of the yield rate in its standard decoction was(43.2±0.1)%,and the mean values of the transfer rates of hesperidin,nobiletin,tangeretin and D-limonene were(19.3±4.9)%,(25.3±4.0)%,(16.6±2.5)%,and(2.2±2.2)%.Conclusion The fingerprints comprehensively reflect the similarity in chemical compositions of Guangchenpi herbs,decoction pieces and standard decoction.The components of standard decoction,including hesperidin,nobiletin and tangeretin,has good positive correlation with that of the herbs and decoction pieces,providing reference for the production,preparation and quality control of Guangchenpi standard decoction,formula granules and other preparations.
