摘 要:目的建立HPLC法同时测定肾衰合剂中7种有效成分的含量。方法色谱柱为Kromasil 100-5-C18(4.6 mm×250 mm,5μm);流动相为乙腈(A)-0.2%甲酸(B)梯度洗脱(0~5 min,12%~14%A;5~25 min,14%~18%A;25~50 min,18%~21%A;50~75 min,21%~30%A;75~85 min,30%~40%A;85~95 min,40%~60%A;95~100 min,60%~12%A);流速1.0 mL·min-1;检测波长260 nm;柱温25℃。结果毛蕊异黄酮葡萄糖苷、新落新妇苷、落新妇苷、橙皮苷、毛蕊异黄酮、芒柄花素、川陈皮素的质量浓度分别在2.01~10.03μg·mL^(-1)(r=0.999)、8.64~43.2μg·mL^(-1)(r=0.995)、5.25~26.25μg·mL^(-1)(r=0.999)、26.67~133.34μg·mL^(-1)(r=0.999)、0.89~4.43μg·mL^(-1)(r=0.999)、0.48~2.42μg·mL^(-1)(r=0.999)、0.48~2.42μg·mL^(-1)(r=0.999)与各自峰面积呈良好的线性关系;精密度、重复性、稳定性(24 h)试验的RSD均未超过3%;平均加样回收率分别为98.81%、105.58%、100.32%、104.68%、106.65%、100.28%、100.44%,RSD均未超过3%。结论该方法简单、重复性好、专属性强,可用于肾衰合剂中多指标成分的含量测定。Objective To establish an HPLC method to determine the content of 7 components in Shenshuai mixture.Methods The chromatographic column was Kromasil 100-5-C18(4.6 mm×250 mm,5μm);the mobile phase was acetonitrile(A)-0.2%formic acid(B)with gradient elution:0~5 min,12%~14%A;5~25 min,14%~18%A;25~50 min,18%~21%A;50~75 min,21%~30%A;75~85 min,30%~40%A;85~95 min,40%~60%A;and 95~100 min,60%~12%A.The flow rate was 1.0 mL·min-1.The detection wavelength was 260 nm and the column temperature was 25℃.Results The mass concentrations of calycosin 7-O-β-Dglucopyranoside,neoastilbin,astilbin,hesperidin,calycosin,formononetin and nobiletin had good linearity at 2.01 to 10.03μg·mL^(-1)(r=0.999),8.64 to 43.2μg·mL^(-1)(r=0.995),5.25 to 26.25μg·mL^(-1)(r=0.999),26.67 to 133.34μg·mL^(-1)(r=0.999),0.89 to 4.43μg·mL^(-1)(r=0.999),0.48 to 2.42μg·mL^(-1)(r=0.999)and 0.48 to 2.42μg·mL^(-1)(r=0.999).The RSDs of precision,repeatability and stability(24 h)were all lower than 3%.The average recoveries were 98.81%,105.58%,100.32%,104.68%,106.65%,100.28%,and 100.44%,and the RSDs were all lower than 3%,respectively.Conclusion This method is simple,repeatitive and exclusive,which can be used to determine multiple components in Shenshuai mixture.
