摘  要:目的以高脂饮食诱导建立肥胖小鼠模型,探索黄芩素(Baicalein,BAI)干预对小鼠糖脂水平、附睾脂肪组织病理形态及肠道微生物结构变化的影响。方法将40只SPF级雄性C57BL/6J小鼠随机分为正常组(ND组)、模型组(HD组)、黄芩素低剂量组(BAI-L组,200mg/kg)和黄芩素高剂量组(BAI-H组,400mg/kg),每组10只。除正常组以普通饲料喂养外,其余各组均以高脂饲料喂养。灌胃12周后,麻醉并眼球采血后处死,留取血清测定空腹血糖(FBG)、胆固醇(TC)和甘油三酯(TG)水平;观察附睾脂肪的病理学改变;收集各组小鼠粪便进行16S rDNA高通量测序。结果HD组小鼠体重、附睾脂肪系数、FBG、TC和TG水平均显著高于ND组(P<0.05,P<0.01);HD组小鼠的附睾脂肪细胞体积明显增大,细胞中可见大量脂肪小泡。与HD组相比,BAI-L、BAI-H组均不同程度的降低肥胖小鼠体重、附睾脂肪系数和糖脂水平,且能明显改善附睾脂肪的病理形态。同时,黄芩素能够在一定程度上调节肥胖小鼠肠道微生物的物种组成,显著增加副萨特氏菌(Parasutterella)的相对丰度,降低布劳特氏菌(Blautia)、颤杆菌克(Oscillibacter)、土拉菌(Tuzzerella)、Lachnospiraceae_UCG-006、Anaerovorax、分枝杆菌属(Negativibacillus)的相对丰度(P<0.05,P<0.01)。结论黄芩素对高脂饮食诱导的肥胖小鼠的体重和糖脂水平具有一定的调控作用,其机制可能与改善小鼠肠道菌群结构丰度及多样性有关。To explore the effects of baicalein(BAI)intervention on glucose and lipid levels,epididymal adipose histopathological changes and intestinal microbial structure changes in obese mice induced by high-fat diet.Methods Forty SPF male C57BL/6J mice were randomly divided into normal group(ND group),model group(HD group),baicalein low-dose group(BAI-L group,200 mg/kg)and baicalein high-dose group(BAI-H group,400 mg/kg),with 10 mice in each group.Except ND group was fed with normal diet,the other groups were fed with high fat diet.After 12 weeks of intragastric administration,the blood was collected from the eyes of anesthetized mice.Serum samples were collected for determining the levels of fasting blood glucose(FBG),cholesterol(TC)and triglyceride(TG).The pathological changes of epididymal fat were observed.Feces were collected from each group for 16S rDNA high-throughput sequencing.Results The body weights,epididymal fat coefficients,the levels of FBG,TC and TG of HD group were significantly higher than those of ND group(P<0.05,P<0.01).The volume of epididymal adipocytes in HD mice increased significantly,and a large number of fat vesicles were observed in the cells.Compared with the HD group,BAI-L and BAI-H groups can reduce the body weights,epididymal fat coefficients and glucose and lipid levels of obese mice to different degrees,and can significantly improve the pathological morphology of epididymal fat.At the same time,baicalein could regulate the species composition of intestinal microorganisms in obese mice to a certain extent,and significantly increased the relative abundance of Parasutterella.The relative abundance of Blautia,Oscillibacter,Tuzzerella,Lachnospiraceae_UCG-006,Anaerovorax and Negativibacillus were decreased(P<0.05,P<0.01).Conclusion Baicalein has certain regulatory effects on body weight and glucose and lipid levels in high-fat diet-induced obese mice,and the mechanism may be related to improving the abundance and diversity of intestinal microbiota structure in mice.
