摘  要:目的观察电针对慢性疲劳综合征(CFS)大鼠行为学和粪便代谢的影响。方法将雄性SD大鼠随机分为空白组(12只)和造模组(30只),造模组采用多因素慢性复合应激法(跑台训练+束缚应激+睡眠干扰+拥挤环境)造模后再随机分为模型组(12只)和电针组(12只)。电针组大鼠取神庭穴透刺百会穴和大椎穴连续干预28天(1次/d)。治疗结束后对各组大鼠进行一般情况(饮食、饮水、体重和高怀林疲劳量表)、旷场实验以及Morris水迷宫实验评价。取各组大鼠的粪便进行非靶向代谢组学研究。结果治疗结束后,与空白组相比,模型组大鼠体重降低(P<0.01),饮食量减少(P<0.05),高怀林疲劳量表评分升高(P<0.01),旷场实验中,穿格次数和进入中央区次数均增多(P<0.01),Morris水迷宫实验中,定位航行到达平台潜伏期延长(P<0.01),空间探索穿越平台次数减少(P<0.01);与模型组相比,电针组大鼠高怀林疲劳量表评分降低(P<0.01),旷场实验中,穿格次数减少(P<0.05),Morris水迷宫实验中,定位航行到达平台潜伏期缩短(P<0.01),空间探索穿越平台次数增加(P<0.01)。与空白组相比,CFS大鼠肠道内有55种表现差异的代谢物,富集到11个代谢通路,与模型组相比,电针组有31种差异代谢物,富集到14条代谢通路。结论电针神庭穴透刺百会穴和大椎穴可以改善CFS模型大鼠的一般状态、缓解焦虑情绪,提高学习记忆能力,并影响CFS大鼠粪便代谢。Objective:To observe the effect of electroacupuncture on behavior and fecal metabolism in rats with chronic fatigue syndrome(CFS).Methods Male SD rats were randomly divided into blank group(n=12)and modeling group(n=30).The modeling group adopted a multi factor chronic composite stress method(treadmill training+restraint stress+sleep disturbance+crowded environment).After modeling,the modeling group was randomly divided into model group(n=12)and electroacupuncture group(n=12).The rats in the electroacupuncture group were treated with electroacupuncture at Shenting(GV24)penetrating Baihui(GV20)and Dazhui(GV14)for 28 days(once a day).The rats in each group were evaluated by general conditions(diet,drinking water,body weight and Gaohuailin fatigue scale),Open Field Test and Morris Water Maze Test.The feces of rats in each group were collected for non-targeted metabolomics study.Results After treatment,compared with the blank group,the rats in the model group had lower body weight(P<0.01),less food intake(P<0.05),higher fatigue scale score(P<0.01)and more times of crossing and central area entering in the Open Field Test(P<0.01),longer latency of searching for a platform in the space navigation experiment(P<0.01)and fewer times of platform crossing in the spatial probe experiment(P<0.01)in the Morris Water Maze Test.Compared with that of the model group,the fatigue scale score of the electroacupuncture group was decreased(P<0.01),the number of crossing in the Open Field Test decreased(P<0.05),the latency of reaching for a platform in the space navigation experiment shortened(P<0.01)and the number of platform crossing in the spatial probe experiment increased(P<0.01)in the Morris Water Maze Test.Compared with the blank group,the CFS rats had 55 different metabolites in feces(P<0.05),which were enriched to 11 metabolic pathways,and compared with the model group,the electroacupuncture group had 31 different metabolites(P<0.05),which were enriched to 14 metabolic pathways.Conclusion Electroacupuncture at Shenting(GV24)pe
