摘  要:细胞焦亡是一种新型细胞程序性死亡,其表现出以NOD样受体蛋白3(NLRP3)为关键靶通路,严重依赖半胱天冬酶(Caspase)家族成员为主要生物学特征,对细胞炎症及免疫调节具有重要意义。肺纤维化是呼吸系统中常见的间质性肺疾病,早期产生不同程度的炎症损伤及修复,后期以肺组织纤维化为主要病理表现。中医药在治疗肺纤维化过程中具有多靶点、多途径等特点。一方面通过阻止细胞炎症介质释放,保护肺组织免受炎症损伤;另一方面,抑制促纤维因子形成,减少细胞外基质及胶原沉积物,阻止肺成纤维细胞向肌成纤维细胞转化。文章就NLRP3/Caspase-1焦亡经典通路机制与PF作用分析,整理、归纳近年来中医药干预细胞焦亡在PF发挥生物活性的研究进展,以期为其深入研究提供参考。Pyrosis is a new type of programmed cell death,which is characterized by NOD-like receptor protein 3(NLRP3)as a key target pathway,heavily dependent on Caspase family members as its main biological characteristics,and has important implications for cellular inflammation and immune regulation.Pulmonary fibrosis is a common interstitial lung disease in the respiratory system,which causes different degrees of inflammatory damage and repair in the early stage,and the main pathological manifestations are lung tissue fibrosis in the later stage.Traditional Chinese medicine has the characteristics of multi-target and multi-pathway in the treatment of pulmonary fibrosis.On the one hand,it protects lung tissue from inflammatory damage by preventing the release of cellular inflammatory mediators.On the other hand,it inhibits the formation of pro-fibrocytokines,reduces extracellular matrix and collagen deposits and prevents the transformation of lung fibroblasts into myofibroblasts.In this paper,the NLRP3/Caspase-1 pyroptosis classical pathway mechanism and the role of pulmonary fibrosis were analyzed,and the research progress on the bioactivity of pulmonary fibrosis induced by the intervention of traditional Chinese medicine in cell pyroptosis in recent years was summarized,in order to provide references for further research.
