摘  要:为探究锝[^(99)Tc]亚甲基二膦酸盐注射液(^(99)Tc-MDP)在大鼠血液中的药代动力学特征,本研究采用电感耦合等离子体-质谱(ICP-MS)直接测定不同时间点^(99)Tc-MDP在大鼠血液中的含量,并用DAS2.1.1软件拟合药-时曲线,得出主要药代动力学参数。结果表明,^(99)Tc-MDP经尾静脉注射给药后,在大鼠血液中的药代动力学性质符合三室模型特征,分布相半衰期T_(1/2α)为0.047 h,表明药物经尾静脉入血后在体内快速分布;消除相半衰期T_(1/2β)、T_(1/2γ)分别为0.113 h、37.349 h,表明药物自机体清除较慢。研究结果可为^(99)Tc-MDP在人体药代动力学方面的研究提供参考。In order to investigate the pharmacokinetic characteristics of technetium [^(99)Tc]methylenediphosphonate injection(^(99)TC-MDP) in the blood of rats,the content of ^(99)Tc-MDP in the blood of rats at different time points was directly determined by inductively coupled plasma-mass spectrometry(ICP-MS),and the drug-time curve was fitted with DAS 2.1.1 software.The main pharmacokinetic parameters were obtained.The results showed that the pharmacokinetic properties of ^(99)Tc-MDP in the blood of rats were consistent with the characteristics of the three-chamber model,and the half-life of the distribution phase T_(1/2α) was 0.047 h,indicating that the drug was rapidly distributed in the body after entering the blood through the tail vein.The elimination phase half-life T_(1/2β) and T_(1/2γ) were 0.113 h and 37.349 h respectively,indicating that the drug was cleared slowly from the body.The results can provide reference for the study of ^(99)Tc-MDP pharmacokinetics in human.
