摘 要:目的:腹膜纤维化是终末期肾病患者腹膜透析治疗引起的一种严重并发症。上皮-间质转化(epithelial-to-mesenchymal transition,EMT)是早期腹膜纤维化的一个促成因素。本研究旨在探讨长链非编码RNA(LncRNA)OIP5-AS1在高糖培养的大鼠腹膜间皮细胞(RPMC)中的作用及相关机制。方法:用OIP5-AS1 siRNA转染RPMC,然后用高糖(high glucose,HG)孵育。应用Transwell法测定RPMC的迁移能力。通过免疫荧光和α-SMA抗体评估RPMC的转分化能力。应用RT-qPCR分析OIP5-AS1、α-SMA、波形蛋白、E-钙黏蛋白、TGF-β_(1)、CTGF和Ⅰ型胶原的基因表达。结果:在HG孵育的RPMC中OIP5-AS1的表达升高。OIP5-AS1沉默降低了HG处理后RPMC的迁移能力,减弱了HG诱导的EMT和转分化。此外,OIP5-AS1沉默降低RPMC分泌的TGF-β_(1),并抑制TGF-β_(1)、CTGF和Ⅰ型胶原的mRNA表达。结论:LncRNA OIP5-AS1通过TGF-β_(1)途径调节EMT、转分化以及迁移。OIP5-AS1是一种新发现的LncRNA,可能与腹膜纤维化的EMT过程有关。Objective:Peritoneal fibrosis(PF)is a severe complication caused by peritoneal dialysis(PD)therapy in patients with end stage renal disease(ESRD),with epithelial-to-mesenchymal transition(EMT)as a contributing factor to PF at the early stage.The current study aims to explore the effect and and related mechanisms of LncRNA OIP5-AS1 in rat peritoneal mesothelial cells(RPMCs)incubated with high glucose.Methods:RPMCs were transfected with OIP5-AS1 siRNA(siOIP5-AS1),followed by incubation with high glucose(HG).Transwell assay was applied to determine the migrative capacity of RPMCs.Transdifferentiation of RPMCs was evaluated by immunofluorescence withα-SMA.Expression of OIP5-AS1,and mRNA ofα-SMA,Vimentin,E-cadherin,TGF-β_(1),CTGF and Collagen I were measured by RT-qPCR.Results:OIP5-AS1 expression was elevated in RPMCs incubated with HG or TGF-β_(1).Silencing of OIP5-AS1 decreased migration RPMCs with HG.In addition,siOIP5-AS1 attenuated the HG-induced transdifferentiation of RPMCs and EMT.Furthermore,siOIP5-AS1 remarkably reduced TGF-β_(1)secretion,and suppressed mRNA expression of TGF-β_(1),CTGF and CollagenⅠ.Conclusion:LncRNA OIP5-AS1 to regulate influence migration,transdifferentiation and EMT of RPMCs via the TGF-β_(1)pathway.OIP5-AS1 is a new identified LncRNA that might be associated with EMT process of peritoneal fibrosis.
