摘  要:目的:研究黄芪甲苷(AS-IV)对糖尿病肾病(DN)模型大鼠肾组织细胞焦亡指标GSDMD及Caspase-1表达的影响。方法:将40只健康SD雄性大鼠随机分为造模组30只和正常组(NC组)10只,造模组腹腔注射链脲佐菌素(STZ)建立糖尿病肾病大鼠模型,随机分为3组:糖尿病肾病组(DN组)、黄芪甲苷组(AS-IV组)和白藜芦醇组(R组)。连续喂养8周,监测大鼠体重、血糖、尿微量白蛋白,8周后处死大鼠经腹主动脉采血并留取肾组织,检测Scr、BUN、TC、TG,肾脏组织行HE染色观察病理变化,免疫组化检测Caspase-1、GSDMD的表达。结果:(1)与NC组相比,DN组大鼠体重减轻,AS-IV组、R组大鼠体重增加(P<0.05),但低于NC组;(2)与NC组相比,DN组大鼠的血糖、尿微量白蛋白、BUN、Scr、TG及TC明显升高(P<0.05);与DN组相比,AS-IV组、R组血糖于第4周无明显变化(P>0.05),第8周明显下降(P<0.05),尿微量白蛋白、BUN、Scr均下降(P<0.05);AS-IV组TG水平降低(P<0.05),R组TG、TC水平均降低(P<0.05);(3)免疫组化显示,与NC组相比,DN组Caspase-1和GSDMD的表达增加(P<0.05);与DN组相比,AS-IV组及R组大鼠肾组织Caspase-1、GSDMD的表达减少(P<0.05)。结论:AS-IV可能通过下调DN大鼠肾组织GSDMD及Caspase-1的表达,改善细胞焦亡,延缓DN的进展。Objective:The study investigated the effect of astragaloside(AS-IV)on the expression of GSDMD and caspase-1 in diabetic nephropathy(DN)rats.Methods:40 healthy SD male rats were randomly divided into two groups,30 model group and 10 normal group(NC group).Model group disposable peritoneal inject streptozotocin(STZ)to establish the rat model of diabetic nephropathy and randomly divided into 3 groups:diabetic nephropathy group(DN group),astragaloside group(AS-IV group)and Resveratrol group(R group).These Rats were fed continuously for 8 weeks,during which parameters such as body weight,blood glucose levels,and urinary microalbumin were monitored.8 weeks later,rats were humanely euthanized.Blood samples were collected via the abdominal aorta,and kidney tissues were harvested.Serum creatinine(Scr),blood urea nitrogen(BUN),total cholesterol(TC),and triglycerides(TG)levels were assessed.Kidney tissue sections underwent HE staining for the observation of pathological changes,and immunohistochemistry was employed to assess the expression of caspase-1 and GSDMD.Results:(1)Relative to the NC group,rats in the DN group exhibited decreased body weight,while those in the AS-IV and R groups demonstrated increased body weight(P<0.05),although it remained lower than that of the NC group.(2)Compared with the NC group,rats in the DN group displayed notably elevated levels of blood glucose,urinary microalbumin,BUN,Scr,TG,and TC(P<0.05).Conversely,rats in the AS-IV and R groups,when compared to the DN group,exhibited no significant alterations in blood glucose levels at the 4th week(P>0.05),but experienced a pronounced reduction at the 8th week(P<0.05).Furthermore,urinary microalbumin,BUN,and Scr exhibited decreased levels in these groups(P<0.05).TG levels saw a reduction in the AS-IV group(P<0.05),while TG and TC levels decreased in the R group(P<0.05).(3)Immunohistochemistry results revealed that,relative to the NC group,the DN group exhibited an augmented expression of caspase-1 and GSDMD(P<0.05).Meanwhile,the AS-IV and R groups d
