摘  要:乳头瘤病毒(PV)是人畜共患的重要病原体。为调查北疆地区马携带的PV类型,本研究对北疆3个地区采集的205份马鼻拭子样品进行高通量测序和宏病毒组学分析,结果显示马匹携带人乳头瘤病毒124型(HPV-124)。根据HPV-124 E6基因序列设计引物,对800份各种临床样品经PCR检测HPV-124 E6基因,PCR结果显示,健康马鼻拭子样品中HPV-124的总阳性率为2.4%(4/168),存在呼吸道疾病的马鼻拭子中HPV-124的阳性率为100%(37/37),表明HPV-124可跨种感染马,且与呼吸道疾病密切相关;在纯血马和伊犁马流产胎儿胎盘、脐带样品中也检测到HPV-124,表明HPV-124可垂直传播,可能与马流产有关。另外,在多个马场工人鼻拭子中均检测到HPV-124,提示马源HPV-124可能来源于人。E6基因序列的同源性分析结果显示,本研究鉴定的2株人源和5株马源HPV-124 E6基因与美国HPV NJ3900株E6基因同源性均为100%,进一步表明马源HPV-124可能来源于人。采用最大似然法绘制E6基因的进化树,结果显示,本研究鉴定的马源HPV-124与人源HPV-124形成独立进化分支,均处于Beta-1进化分支。本研究首次在马体内扩增到HPV-124 E6部分基因序列,它与人源HPV-124同源性达100%,HPV-124可能会跨物种感染马,并导致感染马流产,该研究结果为动物源HPV-124的流行病学研究提供了参考依据。Papillomavirus(PV)is an important pathogen in humans and animals.In order to understand the types of PV carried by horses in Northern Xinjiang,205 nasal swab samples were collected from 3 areas in Northern Xinjiang,and the viral metagenomic*Corresponding author;†Equal contributors sequencing was applied to detect the types of PVs in horses.The result showed that these samples carried human papillomavirus type 124(HPV-124).According to the HPV-124 reference strain,specific primers targeting E6 gene were designed.PCR results showed that the total positive rate of HPV-124 from nasal swabs was 2.4%(4/168),and the positive rate of HPV-124 in nasal swabs of horses with respiratory diseases was 100%(37/37),indicating that HPV-124 could infect horses across species and was closely related to respiratory disease of pure blood horses;HPV-124 was detected in placenta and umbilical cord tissue of thoroughbred horse and Yili horse aborted fetus,indicating that horse-origin HPV-124 could be vertically transmitted and may be associated with horse abortion.HPV-124 was detected in nasal swabs of several horse farm workers,suggesting that the equine HPV-124 may be of human origin.In addition,sequences analysis indicated that the homology of two human-origin and five horse-origin HPV-124 E6 nucleotides identified in this study had 100%homology with the E6 nucleotide of HPV NJ3900 reference strain of America,which further suggested that the horse-origin HPV-124 may be derived from human.The phylogenetic tree based on the E6 gene sequences showed that horse-origin HPV-124 and human HPV-124 were clustered in an independence lineage.In this study,the partial sequence of horse HPV-124 E6 gene was amplified for the first time,which has 100%homology with human-origin HPV-124.HPV-124 may infect horse access species and cause abortion in infected horses.Our results provide fundamental data for the epidemiological study of animal-origin HPV-124.
