摘 要:为拓宽产色素微生物资源,以传统酿造郫县豆瓣为分离源,筛选、鉴定具备产红色素能力的菌株。分别采用硅胶柱层析、紫外可见分光光度、高效液相色谱、傅里叶变换红外光谱、核磁共振波谱、高分辨质谱等方法对菌株产红色素进行纯化和结构表征;同时对菌株产红色素的抑菌活性进行初步研究。结果表明,从郫县豆瓣中筛选获得一株产红色素菌株GH-2,结合形态学、生理生化及16S rRNA基因序列分析等鉴定该菌株为粘质沙雷氏菌(Serratia marcescens)。粘质沙雷氏菌GH-2代谢产红色素结构表征鉴定为2-甲基-3-戊基灵菌红素。S.marcescens GH-2产灵菌红素对副溶血性弧菌(Vibrio parahaemolyticus)具有明显的抑制作用,对金黄色葡萄球菌(Staphylococcus aureus)有一定的抑制作用。To broaden the resources of pigment-producing microorganisms,strains with the ability to produce red pigment were screened and identified from traditional brewing Pixian Douban.The red pigments were purified and characterized by silica gel column chromatography,ultraviolet visible spectrophotometry(UV-Vis),high performance liquid chromatography(HPLC),fourier transform infrared spectrometer(FTIR),nuclear magnetic resonance(NMR),and high resolution mass spectrum(HRMS).Additionally,the antibacterial activity of the red pigment produced by the strain was preliminarily analyzed.Results showed that a red pigment-producing strain GH-2 was screened from Pixian Douban,and it was identified as Serratia marcescens by morphological,physiological,and biochemical characteristics and 16S rRNA gene sequence analysis.The structure of red pigment produced by S.marcescens GH-2 was identified as 2-methyl-3-pentyl prodigiosin.The prodigiosin produced by S.marcescens GH-2 has a significant inhibitory effect on Vibrio parahaemolyticus and has a certain inhibitory effect on Staphylococcus aureus.
