摘　　要：目的建立测定史国公药酒中羟基红花黄色素A含量的高效液相色谱(HPLC)法。方法色谱柱为Kromasil 100-5-C_(18)柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.1%磷酸水溶液(12∶88,V/V),流速为1.0 mL/min,检测波长为403 nm,柱温为30℃,进样量为20μL。结果羟基红花黄色素A质量浓度在2.814~45.025μg/mL范围内与峰面积线性关系良好(r=0.9995,n=5);精密度、稳定性,重复性试验结果的RSD均小于2.0%;平均加样回收率为102.12%,RSD为2.44%(n=6)。结论所建立的方法操作简便,结果准确、稳定,且简化了流动相组成及供试品溶液的制备方法,可用于史国公药酒的质量控制。Objective To establish a high-performance liquid chromatography(HPLC)method for determining the content of hydroxysafflor yellow A in Shiguogong Medicinal Liquor.Methods The chromatographic column was the Kromasil 100-5-C_(18)column(250 mm×4.6 mm,5μm),the mobile phase was acetonitrile-0.1%phosphoric acid aqueous solution(12∶88,V/V),the flow rate was 1.0 mL/min,the detection wavelength was 403 nm,the column temperature was 30℃,and the injection volume was 20μL.Results The linear range of hydroxysafflor yellow A was 2.814-45.025μg/mL(r=0.9995,n=5).The RSDs of precision,stability and repeatability tests were all lower than 2.0%.The average recovery rate of hydroxysafflor yellow A was 102.12%with an RSD of 2.44%(n=6).Conclusion The established method is easy,accurate and stable,which simplifies the composition of mobile phase and the preparation of test solution,and it can be used for the quality control of Shiguogong Medicinal Liquor.
