摘 要:目的 观察穴位埋线调控NLRP3炎性小体信号通路对造影剂肾病大鼠肾功能保护的机制研究。方法 将Sprague Dawley (SD)大鼠随机分为空白组、模型组、穴位埋线组,每组10只。采用吲哚美辛、左旋硝基精氨酸甲酯(L-NAME)、碘海醇注射液建立造影剂肾病大鼠模型。穴位埋线组予穴位简易埋线治疗。比较大鼠肾脏组织的病理变化,比较血清和肾脏组织酶联免疫吸附分析(ELISA)检测BALF中IL-1β、IL-10和TNF-α的水平,Western Blot检测NLRP3通路相关蛋白的表达。结果 与模型组比较,穴位埋线组大鼠肾功能损伤评分降低,HE染色穴位埋线组炎性浸润显著减轻,模型组中肾脏血清及组织中,IL-1β、IL-10和TNF-α显著升高,而穴位埋线组下降明显,同时,Western Blot检测差异均具有统计学意义(P<0.05)。结论 穴位埋线疗法可显著改善造影剂肾病模型大鼠肾脏的炎症,降低IL-1β、IL-10和TNF-α及病理组织学评分,其作用机制可能与调控NLRP3炎性小体信号通路相关。Objective To observe the mechanism of acupoint catgut embedding regulating the NLRP3 inflammasome signaling pathway in protecting renal function in contrast induced nephropathy rats.Method Sprague Dawley(SD)rats were randomly divided into blank group,model group and acupoint embedding group,with 10 rats in each group.Indomethacin,L-nitroarginine methyl ester(L-NAME)and iohexol injection were used to establish the rat model of contrast nephropathy.The acupoint embedding group was given simple embedding treatment.The pathological changes of rat kidney tissue were compared.The levels of IL-1β,IL-10 and TNF-αin BALF in in serum and kidney tissue detected by enzyme-linked immunosorbent assay(ELISA)were compared.Western blot was used to detect the expressions of NLRP3 pathway related proteins.Results Compared with the model group,the score of renal function damage in the acupoint catgut embedding group was reduced,the inflammatory infiltration in the acupoint catgut embedding group was significantly reduced by HE staining,and the levels of IL-1β,IL-10 and TNF-αin the serum and tissue of the kidney in the model group were significantly increased,while in the acupoint catgut embedding group,there was a significant decrease.At the same time,the differences in Western Blot detection were statistically significant(P<0.05).Conclusion Acupoint catgut therapy can significantly improve kidney inflammation in contrast nephropathy model rats,and reduce the levels of IL-1β,IL-10 TNF-αand histopathological scores.The mechanism of action may be related to the regulation of NLRP3 inflammasome signaling pathway.
