摘 要:目的探讨淫羊藿苷对大鼠正畸牙移动后保持期牙周组织的改建和核因子κB受体活化因子配体(RANKL)表达的影响。方法采用40只6周龄雄性SD大鼠建立正畸牙移动模型,在牙移动21 d后进入保持期,随机将大鼠分为4组,淫羊藿苷组、对照组、淫羊藿苷+保持丝组、保持丝对照组,每组10只。淫羊藿苷组和淫羊藿苷+保持丝组通过灌胃方式给予大鼠淫羊藿苷20 mg/(kg·d),对照组和保持丝对照组灌注等量生理盐水。在实验第1、3、7、14、21天处死大鼠,采集其上颌骨样本进行检测,确定出上颌第一磨牙远中移动复发距离。全部样本脱钙后制成切片,给予苏木精-伊红染色法(HE)染色和免疫组化染色,观察RANKL表达的变化。结果淫羊藿苷及淫羊藿苷+保持丝组在第7、14和21天,牙移动复发距离小于对照组。免疫组化显示,在保持阶段给药期间,从第1天至21天,淫羊藿苷+保持丝组牙周膜组织中RANKL表达低于对照组,差异有统计学意义(P<0.05)。结论淫羊藿苷影响牙槽骨的改建,可以降低破骨细胞活性,有利于正畸牙移动到位后的保持。Objective This study was designed to investigate the effect of icariin on the periodontal tissue reconstruction and the expression of receptor activator of nuclear factor-kappa B ligand(RANKL)in rats during the retention stage after orthodontic tooth movement.Methods The orthodontic tooth movement model was established in 406-week-old male Sprague-Dawley(SD)rats.The rats were randomly divided into 4 groups(icariin group,control group,icariin+maintainer group and maintainer group),with 10 rats in each group.During the retention period(after 21 days of active force application),rats in the experimental group were given a gavage of icariin 20 mg/(kg·d),while rats in the control group were given the same amount of 0.9%sodium chloride everyday.The rats were sacrificed on the 1st,3rd,7th,14th and 21st day of the retention period.The maxillary bone samples were collected for detection.The maxillary first molar distal moving recurrence distance was determined.After drug infusion,the collected tissue specimens were stained with hematoxylin-eosin(HE)to observe the periodontal reconstruction,and the expression of RANKL was detected by immunohistochemical staining.Results The distance of tooth movement recurrence in the icariin and the icariin+maintainer groups was significantly smaller than that in the control group on the 7th,14th and 21st day during the retention period(P<0.05).Immunohistochemistry showed that the expression of RANKL in the periodontal tissue of the icariin+maintainer group was lower than that of the control group from the 1st day to the 21st day during the retention period,and the difference was significant(P<0.05).Conclusion Icariin affects alveolar bone remodeling by inhibiting osteoclasts activity,which is beneficial to the retention of orthodontic teeth after moving into position.
