摘 要:为了明确云南省某乌骨鸡种鸡场鸡白痢沙门氏菌的分子生物学特性,试验收集了90份18~21日龄的未出壳死胚样品和100份下痢鸡的泄殖腔棉拭子,采用沙门氏菌属血清诊断试剂盒和PCR方法进行病原菌的分离鉴定,并对分离菌进行MLST分型、药敏试验、毒力基因和耐药基因检测。结果表明:在90份未出壳死胚样品中共分离到80株鸡白痢沙门氏菌(88.89%,80/90),而泄殖腔棉拭子样品中未分离到鸡白痢沙门氏菌。80株鸡白痢沙门氏菌均为ST92型(100%,80/80);均携带invA、hilA、sefA、sipA、sipC、sopB、sopE、spvC、ssaR、ssrA、stn P1等11种毒力基因,均未检出毒力基因pef A和rck;对红霉素的耐药率最高,为93.75%(75/80),而对氧氟沙星、庆大霉素、氟苯尼考、多黏菌素B、多西环素、加替沙星、氨苄西林、阿莫西林、头孢西丁、头孢他啶、链霉素、四环素和复方新诺明均敏感;均未检出耐药基因,与耐药表型一致。说明从云南省某乌骨鸡种鸡场分离到的鸡白痢沙门氏菌来源单一,80株鸡白痢沙门氏菌均携带11种毒力基因,提示其毒力较强;除了红霉素外,对其他药物均敏感,提示该乌骨鸡种鸡场用药相对安全。In order to clarify molecular biological characterization of Salmonella pullorum from a breeding farm of Black-Boned chicken in Yunnan Province,in the test,ninety samples of unembryonic dead embryos from 18 to 21 days of age and 100 cloacal swabs of chickens with dysentery were collected;Salmonella sero-diagnostic kit and PCR method were used for the isolation and identification of pathogenic bacteria;and the isolates were subjected to carry out MLST typing,drug susceptibility test,and detection of virulence genes and drug resistance genes.The results showed that a total of 80 strains of Salmonella pullorum(88.89%,80/90)were isolated from 90 samples of unembryonated dead embryos;whereas Salmonella pullorum was not isolated from cloacal swab samples.All 80 strains of Salmonella pullorum were ST92(100%,80/80);all carried invA,hilA,sefA,sipA,sipC,sopB,sopE,spvC,ssaR,ssrA,and stnP1,for a total of 11 virulence genes;none of them had detectable virulence genes pefA and rck.The highest resistance rate was 93.75%(75/80)to erythromycin;whereas it was sensitive to ofloxacin,gentamicin,florfenicol,polymyxin B,doxycycline,gatifloxacin,ampicillin,amoxicillin,cefoxitin,ceftazidime,streptomycin,tetracycline,and cotrimoxazole;all of them did not have any detectable resistance genes,which was in line with the resistance phenotype.The results indicated that Salmonella pullorum isolated from a breeding farm in Yunnan Province was from a single source;all 80 strains of Salmonella pullorum carried 11 virulence genes,suggesting strong virulence;they were sensitive to all drugs except erythromycin,suggesting that it was relatively safe to use drugs in this farm.
