摘 要:[目的]建立中药材赤瓟不同药用部位(块根和果实)中原儿茶醛和原儿茶酸的含量测定方法。[方法]采用高效液相色谱法(HPLC)测定赤瓟(块根和果实)中原儿茶醛和原儿茶酸的含量。色谱条件:Discovery C_(18)柱(4.6 mm×250 mm,5μm),流动相为甲醇-5%磷酸水溶液进行梯度洗脱(0~40 min,甲醇:20%~100%),紫外检测波长:280 nm,流速:1 m L/min。[结果]原儿茶醛的含量在0.003~0.052 mg/m L范围内具有良好的线性关系(r=1.000 0),平均回收率为100.6%,RSD为1.27%;原儿茶酸的含量在0.002~0.042 mg/m L范围内具良好的线性关系(r=1.000 0),平均回收率为100.6%,RSD为1.76%。[结论]该测定方法操作简单、结果可靠、重复性好、定量准确,可用于赤瓟(块根和果实)中原儿茶醛和原儿茶酸的含量测定。[Objective]To establish an assay method to determine the content of protocatechuic aldehyde and protocatechuic acid in the roots and fruits of Thladiantha dubia.[Method]Protocatechuic aldehyde and protocatechuic acid were separated and analyzed by Discovery C_(18) column(4.6 mm×250 mm,5μm),mobile phase was methanol-5%phosphoric acid(0-40 min,methanol:20%~100%)with gradient elution at the flow rate of 1 mL/min and the detection wavelength was set at 280 nm.[Result]Protocatechuic aldehyde had good linear relationship between 0.003-0.052 mg/mL(r=1.0000),the average recovery was 100.6%and RSD was 1.27%;protocatechuic acid had good linear relationship between 0.002-0.042 mg/mL(r=1.0000),the average recovery was 100.6%and RSD was 1.76%.[Conclusion]The method is simple,sensitive and reliable to carry out and control the quality of protocatechuic aldehyde and protocatechuic acid in the roots and fruits of T.dubia.
