摘 要:FtsZ蛋白作为微管蛋白同源物,常通过自我组装成一个收缩的细胞骨架元素“Z”环,影响质体分裂。但是,黄瓜FtsZ蛋白的研究尚未见报道,其功能尚不明确。本文利用生物信息学方法在黄瓜中鉴定到了3个FtsZ蛋白,并将它们分为2组。序列分析发现它们的长度在4493~6264bp之间,含有6~7个外显子,氨基酸长度为421~488AA,分布在3号或6号染色体上;亚细胞定位预测结果显示,黄瓜FtsZ定位于叶绿体或细胞质;RNA-seq分析发现黄瓜FtsZs在茎、叶、雌花和子房表达较高,荧光定量PCR结果显示黄瓜CsFtsZ2-1基因在雄花中的表达量也较高,表明黄瓜FtsZs基因在叶、花和子房生长发育中发挥功能;此外,在胁迫响应过程中,CsFtsZ2-1基因可响应盐胁迫,CsFtsZ1、CsFtsZ2-1和CsFtsZ2-2可响应冷胁迫,CsFtsZ1和CsFtsZ2-1可响应白粉病病原菌侵染。本文章结果为进一步研究FtsZs基因在黄瓜中的作用和机制提供了参考和依据。FtsZ protein,as a homolog of tubulin,often assembles into a contractile cytoskeletal element known as the“Z”ring,influencing plastid division.However,research on cucumber FtsZ proteins has not been reported,and its function is unclear.In this paper,we identified three FtsZ proteins in cucumber by bioinformatics methods and classified them into two groups.Sequence analysis revealed that their lengths range from 4493 to 6264 bp,containing 6 to 7 exons,with amino acid lengths of 421 to 488 AA,located on chromosome 3 or 6;subcellular localization predictions indicated that cucumber FtsZ is located in the chloroplast or cytoplasm;RNA-seq analysis found that cucumber FtsZs are highly expressed in stem,leaf,female flower,and ovary,and fluorescence quantitative PCR results showed that the expression level of cucumber CsFtsZ2-1 gene is also high in male flowers,indicating that cucumber FtsZs genes play a role in the growth and development of leaves,flowers,and ovaries;additionally,in the process of responding to coercion,the CsFtsZ2-1 gene can respond to salt stress,CsFtsZ1,CsFtsZ2-1,and CsFtsZ2-2 can respond to cold stress,and CsFtsZ1 and CsFtsZ2-1 can respond to powdery mildew pathogen infection.The research results provide a reference and basis for further study of the role and mechanism of FtsZs genes in cucumber.
