摘  要:KR(山奈酚-3-O-芸香糖苷)具有药理高附加值。采用水热酸控醇提法提制桑叶中KR,优化获得KR浸提适宜参数,构建动力学模型,探究KR的传质溶浸过程。结果表明:投加柠檬酸质量浓度0.050 g/L,桑叶与45%的乙醇溶液质量体积比为1∶20 g/mL,80℃水热反应釜中处理40 min,2.00 g桑叶可溶浸KR 16.09 mg/g,是自动中药煲水热浸提KR(0.41 mg/g)的40倍;SEM(扫描电镜)显示水热酸控反应后桑叶表面孔隙明显增多增大,FT-IR(傅里叶变换红外光谱)表明水热酸控反应有利于桑叶C-O、C-H等键水裂解,KR溶浸满足一级动力学模型,溶出速率为-3.45 min-1,为桑叶高值化开发提供理论依据。KR(kaempferol-3-O-rutinoside)possesses high pharmacological value.The hydrothermal acid-controlled alcohol extraction method was employed to extract KR from mulberry leaves.The parameters for KR extraction were optimized,and a kinetic model was developed to investigate the mass transfer solubilization and leaching process of KR.The results reveal that the addition mass concentration of citric acid is 0.050 g/L,along with the mass volume ratio of mulberry leaves to 45%ethanol solution of 1:20 g/mL,and after treatment in hydrothermal reactor at 80℃for 40 min,2.00 g of mulberry leaves can be soluble with KR 16.09 mg/g.This is 40 times higher than the content achieved through hydrothermal leaching of KR in an automatic traditional Chinese medicine boiler(0.41 mg/g).SEM(scanning electron microscope)analysis demonstrates a significant increase in surface pores of mulberry leaves after the hydrothermal acid-controlled reaction.Furthermore,FT-IR(Fourier-transform infrared spectroscopy)indicats that the hydrothermal acid-controlled reaction facilitats the cleavage of C—O and C—H bonds in the mulberry leaves,and the dissolution of KR follows a first-order kinetic model with a dissolution rate of-3.45 min-1.These results provide a theoretical basis for the development of mulberry leaves with high value.
