摘 要:目的 研究维生素D衍生物卡泊三醇对小鼠斑秃的缓解情况及对相关炎症因子的影响。方法 构建小鼠斑秃模型,将维生素D衍生物卡泊三醇均匀涂抹在小鼠斑秃区域后,观察小鼠斑秃缓解情况;同时,通过免疫荧光染色技术检测斑秃区域皮肤组织CD4^(+)、CD8^(+) T细胞和肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)等细胞因子情况。结果 卡泊三醇处理后小鼠斑秃完全消失,治疗有效率达100%;小鼠毛囊细胞周围与炎症反应相关的CD4^(+)和CD8^(+)T细胞数量降低,TNF-α、IL-6等炎症细胞因子分泌水平下降(P<0.05)。结论 卡泊三醇可通过抑制小鼠毛囊细胞周围与炎症反应相关的CD4^(+)和CD8^(+)T细胞数量以及TNF-α、IL-6等炎症细胞因子分泌而改善小鼠斑秃。Aim To investigate the efficacy of Calciportriol,a vitamin D derivative,on mice with alopecia areata and related inflammatory factors.Methods Mouse models of alopecia areata were constructed.Then,the vitamin D derivative calcipotriol was evenly smeared on the alopecia areata area of the mice.The efficacy of alopecia areata in the mice was observed.Furthermore,immunofluorescence staining was used to detect CD4~+,CD8~+ T cells and related cytokines,such as TNF-α and IL-6,in alopecia areata areas.Results Alopecia areata disappeared completely after treatment with Calciportriol,and the effective rate was 100%.The number of CD4~+ and CD8~+T cells associated with inflammation around mouse hair follicle cells decreased,accompanying with suppressed secretion of inflammatory cytokines such as TNF-α and IL-6(P<0.05).Conclusion Carpotriol can significantly improve alopecia areata in mice by inhibiting the number of CD4~+ and CD8~+T cells and the secretion of TNF-α,IL-6 and other inflammatory cytokines around hair follicle cells.
