摘 要:目的 探讨山银花黄酮对氧化低密度脂蛋白(oxidized low-density lipoprotein,Ox-LDL)诱导的血管平滑肌细胞(vascular smooth muscle cells,VSMCs)增殖和迁移的影响和可能机制。方法 采用MTT、划痕愈合、Transwell实验检测山银花黄酮(62.5、125、250μg/mL)对Ox-LDL诱导的VSMCs增殖和迁移的影响。蛋白质印记法检测E-钙黏蛋白和N-钙黏蛋白表达。RT-qPCR检测miR-134-5p表达。将miR-134-5p抑制物、miR-134-5p模拟物分别转染VSMCs,检测抑制miR-134-5p表达或过表达miR-134-5p联合山银花黄酮对Ox-LDL诱导的VSMCs增殖和迁移的影响。结果 Ox-LDL处理后VSMCs活力、迁移距离、迁移数量、N-钙黏蛋白、miR-134-5p显著增加,E-钙黏蛋白显著减少。山银花黄酮显著降低Ox-LDL诱导VSMCs的活力、迁移距离、迁移数量以及N-钙黏蛋白、miR-134-5p表达,增加E-钙黏蛋白表达。抑制miR-134-5p显著降低Ox-LDL诱导VSMCs的活力、迁移距离、迁移数量以及N-钙黏蛋白表达,增加E-钙黏蛋白表达。过表达miR-134-5p显著减弱山银花黄酮处理对Ox-LDL诱导VSMCs活力、迁移以及N-钙黏蛋白、E-钙黏蛋白表达的影响。结论 山银花黄酮可能通过下调miR-134-5p表达来抑制Ox-LDL诱导的VSMCs增殖和迁移。Objective To investigate the effect and possible mechanism of lonicera flavonoids on the proliferation and migration of oxidized low-density lipoprotein(Ox-LDL)-induced vascular smooth muscle cells(VSMCs).Methods Effects of lonicera flavonoids(62.5,125,250 μg/mL) on the proliferation and migration of Ox-LDL-induced VSMCs were calculated by MTT experiment,scratch healing experiment,and Transwell experiment.Western blotting was used to detect E-cadherin and N-cadherin expression.RT-qPCR was employed to detect miR-134-5p expression.The influence of miR-134-5p inhibition or miR-134-5p overexpression combined with lonicera flavonoids on the proliferation and migration of Ox-LDL-induced VSMCs were assessed using the above methods.ResultsThe activity,migration distance,migration number,N-cadherin and miR-134-5p of VSMCs were significantly increased after Ox-LDL treatment,while E-cadherin was significantly decreased.The treatment of lonicera flavonoids significantly reduced the activity,migration distance,migration number,and N-cadherin expression as well as miR-134-5p expression of Ox-LDL-induced VSMCs,while increased the expression of E-cadherin.miR-134-5p inhibition significantly reduced the activity,migration distance,migration number and N-cadherin protein expression of Ox-LDL-induced VSMCs,while increased E-cadherin expression.miR-134-5p overexpression significantly weakened the effects of lonicera flavonoids treatment on the activity and migration,as well as N-cadherin and E-cadherin expression of Ox-LDL-induced VSMCs.Conclusion Lonicera flavonoids may inhibit the proliferation and migration of Ox-LDL-induced VSMCs by down-regulating miR-134-5p.
