摘　　要：肺动脉高压是一类以肺血管结构和功能改变、肺动脉压力增加导致右心衰竭为特征的进展性、致命性疾病。哺乳动物雷帕霉素靶蛋白(mTOR)是调控细胞增殖、代谢和生存的关键因子,结合不同辅助蛋白分别组成mTOR复合物1(mTORC1)和mTOR复合物2(mTORC2),在肺动脉高压多个病理环节中发挥核心调控作用,与肺血管重构和右心功能障碍的发生发展密切相关,mTORC1可能是具有前景的肺动脉高压治疗靶点。本文对mTOR信号通路在肺动脉高压中的作用及针对这一信号通路的靶向治疗的研究进展进行综述。Pulmonary hypertension(PH) is a kind of progressive and fatal disease characterized by changes in the structure and function of pulmonary vessels and right heart failure caused by increased pulmonary artery pressure.Mammalian rapamycin target protein(mTOR) is a key node in regulating cell proliferation,metabolism and survival.It combines different subunits to form mTOR complex 1(mTORC1) and mTOR complex 2(mTORC2) respectively,which play vital roles in multiple pathological stages of PH and are closely associated with the occurrence and development of pulmonary vascular remodeling and right ventricular dysfunction.mTORC1 may be a promising therapeutic target for PH.In this review,we summarized the roles of mTOR signal pathway in PH and the latest research progress of targeted therapy of this signal pathway in PH.
