摘 要:目的:探讨芩百清肺浓缩丸(芩百)对感染后咳嗽(PIC)大鼠NOD样受体热蛋白结构域相关蛋白3/半胱氨酸天冬氨酸蛋白水解酶1/白细胞介素1β(NLRP3/Caspase-1/IL-1β)信号通路的调节作用。方法:40只SD大鼠随机分为正常组、模型组、阳性对照组(阿斯美)及芩百清肺浓缩丸(芩百)组,每组10只。采用烟熏联合LPS滴鼻法及辣椒素雾化吸入法构建PIC模型。于造模后第18天开始灌胃给药,芩百组给予芩百清肺浓缩丸水溶液1.65 g/kg,阳性对照组给予阿斯美25.11 mg/kg,每日1次,连续给药10 d。末次干预后,ELISA检测BALF中IL-1β含量,、Western blot检测肺组织NLRP3、Caspase-1蛋白表达。结果:与正常组相比,模型组BALF中IL-1β含量以及肺组织NLRP3、Caspase-1蛋白表达均显著升高(P<0.05);与模型组相比,阳性对照组和芩百组上述指标均显著降低(P<0.05);芩百组BALF中IL-1β水平和肺组织NLRP3蛋白表达均显著低于阳性对照组(P<0.05)。结论:芩百清肺浓缩丸能够显著减轻PIC大鼠的气道炎症和气道高反应性,其作用机制可能与抑制NLRP3/Caspase-1/IL-1β信号通路有关。Objective:To observe the effects of Qinbai Qingfei Concentrated Pills(Qinbai)on NLRP3/Caspase-1/IL-1βsignaling pathway in rats with post-infection cough(PIC).Methods:40 healthy male SD rats were randomly divided into normal group,model group,positive control group(Asmeton)and Qinbai Qingfei Concentrated Pills group(Qinbai),with 10 rats in each group.The PIC model was constructed by smoking combined with LPS nasal drip and aerosol inhalation of capsaicin.After modeling(day 18),intragastric administration was started.The Qinbai group was given Qinbai Qingfei Concentrated Pills solution 1.65 g/kg,and the positive control group was given Asmeton 25.11 mg/kg,each group was administered once daily for 10 days.After the last intervention,the level of IL-1βin BALF was detected by ELISA;the protein expressions of NLRP3 and Caspase-1 in lung tissues were detected by Western blot.Results:Compared with normal group,the levels of IL-1βin BALF and the protein expressions of NLRP3 and Caspase-1 in lung tissues in the model group were significantly increased(P<0.05).Compared with model group,those in Qinbai group and positive control group were significantly decreased(P<0.05).Moreover,the levels of IL-1βand the levels of NLRP3 protein expression in lung tissue in Qinbai group were significantly lower than those in control group(P<0.05).Conclusion:Qinbai Qingfei concentrated pill can significantly reduce airway inflammation and airway hyperreactivity in rats with PIC,and its mechanism may be related to inhibiting NLRP3/Caspase-1/IL-1βpathway.
