摘  要:目的:建立一测多评(QAMS)法同时测定华山参中东莨菪碱、山莨菪碱、东莨菪苷、莨菪碱、东莨菪内酯等成分的含量。方法:采用Agilent 5 TC-C_(18)(250 mm×4.6 mm,5μm)色谱柱,以甲醇-0.1%磷酸水溶液为流动相,梯度洗脱,流速1.0 mL·min^(-1),检测波长为210 nm和344 nm,柱温30℃,进样量10μL。以山莨菪碱为内参物,计算东莨菪碱、东莨菪苷、莨菪碱、东莨菪内酯的相对校正因子,测定其含量,并与外标法(ESM)测定结果进行比较。结果:5个成分在各自范围内线性关系良好(r=1.000),东莨菪碱、东莨菪苷、莨菪碱、东莨菪内酯的相对校正因子分别为0.9518、0.5626、0.8304、0.4339,重复性较好。QAMS法计算所得10个产地华山参中东莨菪碱、山莨菪碱、东莨菪苷、莨菪碱、东莨菪内酯的含量与ESM实测值无显著性差异。结论:该方法准确稳定,重复性好,可用于华山参的质量控制。Objective:To establish a quantitative analysis of multi-components by single marker(QAMS)method to simultaneously determine the contents of scopolamine,anisodamine,scopolin,hyoscyamine and scopoletin in Physochlaina infundibularis Kuang.Methods:Agilent 5 TC-C_(18)(250 mm×4.6 mm,5μm)column was used with methanol-0.1%phosphoric acid aqueous solution as mobile phase for gradient elution at a flow rate of 1.0 mL·min^(-1).The detection wavelengths were 210 nm and 344 nm,the column temperature was 30°C,and the injection volume was 10μL.The relative correction factors for scopolamine,scopolin,hyoscyamine and scopoletin and anisodamine were calculated by using anisodamine as the internal reference substance.The contents were determined and compared with the results of external standard method(ESM).Results:The five components had good linear relationship in their respective ranges(r=1.000).The relative correction factors of scopolamine,anisodamine,scopolin,hyoscyamine and scopoletin were 0.9518,0.5626,0.8304 and 0.4339,respectively,with good repeatability.The contents of scopolamine,anisodamine,scopolin,hyoscyamine and scopoletin in P.infundibularis from 10 habitats calculated by QAMS method were not significantly different from those determined by ESM.Conclusion:The method is accurate,stable and reproducible,and can be used for the quality control of P.infundibularis.
