摘  要:目的研究没食子水提物对溃疡性结肠炎(UC)的疗效及其作用机制。方法采用2,4,6-三硝基苯磺酸建立UC大鼠模型,将UC模型大鼠随机均分为模型组、美沙拉嗪组和没食子水提物低、中、高剂量组,另设正常组;正常组和模型组ig饮用水,其余各组分别ig相应浓度的药物,每日1次,连续15 d。给药过程中定期进行疾病活动指数(DAI)评分;大鼠解剖后进行Luk评分;用血细胞分析仪进行细胞分类检测;ELISA法检测结肠组织中相关炎症因子的表达水平;HE染色法观察结肠组织的病理学变化;RT-PCR检测结肠组织中转化生长因子、三叶因子3、β-防御素2的mRNA转录水平。结果没食子水提物可显著降低UC大鼠的DAI、Luk评分,调控炎症因子及修复因子的表达水平,改善UC大鼠的结肠病变程度。结论没食子水提物可能是通过抑制免疫炎症反应、调控促炎因子及修复因子的表达水平,促进结肠溃疡愈合,从而发挥抗UC的作用。OBJECTIVE To explore the effects and mechanism of nutgall water extracts(NWE)on ulcerative colitis(UC).METHODS The UC rat model was established using 2,4,6-trinitrobenzene sulfonic acid.UC model rats were randomly divided into model group,Mesalazine group,and low,medium,and high dose of NWE groups.A normal control group was previously established.The normal group rats were given water by gavage,and each treatment group was given corresponding concentrations of drugs by gavage once a day for 15 consecutive days.Disease activity index(DAI)scoring was performed regularly during the administration process.After dissection,Luk scoring was performed.Cell classification was performed using a blood cell analyzer.Expression levels of related inflammatory factors in colon tissue were detected by ELISA,and the pathological changes in colon tissue were observed by HE staining.The mRNA transcription levels of transforming growth factor,intestinal trefoil factor 3 andβ-defensin 2 in colon tissues were detected by RT-PCR.RESULTS NWE significantly reduced DAI and Luk scores in UC rats,regulated the expression levels of inflammatory and repair factors,and improved the degree of colon lesions in UC rats.CONCLUSION NWE may promote the healing of colon ulcers by inhibiting immune inflammatory response,regulating the expression levels of pro-inflammatory and repair factors,and thus exerting anti-ulcerative colitis effects.
