蛇床子素下调miR-143-3p抑制TGF-β1诱导的肺成纤维细胞增殖和胶原合成的实验研究

摘  要:目的 观察蛇床子素对小鼠肺成纤维细胞(MLF)增殖和胶原合成的影响并探讨其可能的分子机制。方法 体外培养MLF,将其分为对照组、TGF-β1组、低剂量蛇床子素组、中剂量蛇床子素组、高剂量蛇床子素组、anti-miR-NC组、anti-miR-143-3p组、高剂量蛇床子素+miR-NC组、高剂量蛇床子素+miR-143-3p组;RTqPCR检测微小核糖核酸-143-3p(miR-143-3p)的表达水平;MTT检测细胞活力;Western blotting检测蛋白表达。结果 与对照组比较,TGF-β1组MLF的miR-143-3p表达水平、增殖和胶原合成能力显著升高,细胞周期蛋白D1(CyclinD1)、增殖细胞核抗原(PCNA)、Ⅰ型胶原蛋白(ColⅠ)和Ⅲ型胶原蛋白(ColⅢ)表达水平显著升高(P <0.05);与TGF-β1组比较,中、高剂量蛇床子素组MLF的miR-143-3p表达水平、增殖和胶原合成能力显著降低,CyclinD1、PCNA、ColⅠ和ColⅢ表达水平显著降低(P <0.05);下调miR-143-3p显著降低MLF的增殖和胶原合成能力,CyclinD1、PCNA、ColⅠ和ColⅢ表达水平显著降低(P <0.05);上调miR-143-3p逆转了蛇床子素对MLF增殖和胶原合成的影响(P <0.05)。结论 蛇床子素可能通过下调miR-143-3p表达抑制小鼠MLF的增殖和胶原合成。Objective:To investigate the effects of osthole on mouse lung fibroblasts MLF proliferation and col-lagen synthesis and its possible molecular mechanism.Methods:MLF were cultured in vitro and divided into con-trol group,TGF-β1 group,low dose osthole group,medium dose osthole group,high dose osthole group,anti-miR-NC group,anti-miR-143-3p group,high dose osthole+miR-NC group,high dose osthole+miR-143-3p group.RT-qPCR was used to detect the expressions of miR-143-3p.MTT was used to detect cell activity.Western blotting was used to detect protein expression.Results:Compared with the control group,the expression level of miR-143-3p,proliferation and collagen synthesis ability of MLF in TGF-β1 group were significantly increased,and the ex-pression levels of CyclinD1,PCNA,ColⅠand ColⅢwere significantly increased(P<0.05).Compared with TGF-β1 group,medium and high doses of osthole significantly decreased the expression level of miR-143-3p,prolifera-tion and collagen synthesis ability of MLF,and the expression levels of CyclinD1,PCNA,ColⅠand ColⅢwere significantly decreased(P<0.05).Downregulation of miR-143-3p significantly decreased the proliferation and col-lagen synthesis ability of MLF,and the expression levels of CyclinD1,PCNA,ColⅠand ColⅢwere significantly decreased(P<0.05).The up-regulation of miR-143-3p reversed the effects of osthole on MLF proliferation and collagen synthesis(P<0.05).Conclusion:Osthole may inhibit the proliferation and collagen synthesis of mouse lung fibroblasts MLF by down-regulating the expression of miR-143-3p.
