程氏蠲痹汤对佐剂性关节炎大鼠体内氧化应激的影响

摘　　要：为探究程氏蠲痹汤(Cheng Shi Juan Bi Tang,CSJBT)对佐剂性关节炎(adjuvant arthritis,AA)大鼠体内氧化应激的影响,文章选用弗氏完全佐剂(complete Freund’s adjuvant,CFA)诱导SD大鼠建立关节炎模型。动物随机分为正常组、模型组、雷公藤多苷组(glycosides of Tripterygium wilfordii,GTW)(剂量0.01 g/kg)和CSJBT组(剂量分别为2.55、5.10、10.20 g/kg)。大鼠在造模后的第15天灌胃给药,连续治疗2周后,考察CSJBT对AA大鼠足爪肿胀度、体质量及氧化应激相关指标的影响,并采用HE染色法观察其对AA大鼠关节组织病理学变化的影响。结果显示CSJBT可缓解AA大鼠的关节肿胀,增加其体质量,改善关节滑膜组织病理学变化,减少滑膜组织中活性氧(reactive oxygen species,ROS)的产生以及血清中诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS)和丙二醛(MDA)的水平,而升高超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)及谷胱甘肽(GSH)的水平。结果表明CSJBT对AA大鼠体内的氧化应激反应有较好的抑制作用。To investigate the effects of Cheng Shi Juan Bi Tang(CSJBT)on oxidative stress in adjuvant arthritis(AA)rats,complete Freund’s adjuvant(CFA)was used to induce SD rats to establish arthritis model.The animals were randomly divided into normal group,model group,glycosides of Tripterygium wilfordii(GTW)group(0.01 g/kg)and CSJBT groups(2.55,5.10,and 10.20 g/kg,respectively).The rats were given intragastric administration on the 15th day after modelling for two weeks.The effects of CSJBT on paw swelling degree,body weight and oxidative stress related indexes of AA rats were investigated,and its effects on joint histopathological changes of AA rats were observed by haematoxylin and eosin(HE)staining method.The results showed that CSJBT could alleviate joint swelling,increase body weight,and improve synovial histopathological changes of AA rats.And CSJBT could reduce the production of reactive oxygen species(ROS)in synovial tissues and the levels of inducible nitric oxide synthase(iNOS)and malondialdehyde(MDA)in serum,while increasing the levels of superoxide dismutase(SOD),catalase(CAT),glutathione peroxidase(GSH-Px)and glutathione(GSH)of AA rats.These results indicated that CSJBT had a good inhibitory effect on the oxidative stress response in AA rats.
