白及多糖对生殖氧化损伤模型小鼠的保护作用

摘  要:目的探究白及多糖对生殖损伤模型小鼠生殖功能损伤的保护作用。方法将雄性小鼠随机分为6组,每组6只,分别是空白对照组、生精胶囊对照组、模型对照组以及白及多糖低、中、高剂量组,除空白对照组外,其余5组采用奥硝唑(ornidazole,ORN)500 mg/kg灌胃小鼠21 d建立生殖损伤小鼠模型,再灌胃30 d相应药物,检测小鼠的体重变化;测定脏器指数、精子质量,附睾氧化损伤水平,并观察小鼠睾丸组织病理结构。结果与模型组相比,中剂量组的睾丸指数与附睾指数显著升高(P<0.05),中剂量组与高剂量组的精子活率显著增加(P<0.05),超氧化物歧化酶(SOD)显著升高;观察小鼠睾丸组织病理结构,发现白及多糖处理的中、高剂量组生精细胞数量明显增多。结论白及多糖可提高ORN诱导的生殖损伤小鼠模型的睾丸指数、附睾指数和精子细胞数目与活率,对生殖系统氧化损伤具有一定的改善作用。Objective To investigate the protective effect of Bletilla striata polysaccharides on the reproductive dysfunction of a mice model induced by ornidazole(ORN).Methods Male mice were randomly divided into six groups,each containing six mice:a blank control group,a reference capsule control group,a model control group,and three Bletilla striata polysaccharide dose groups(low,medium,and high).Except for the blank control group,all other mice were administered ORN at a dose of 500 mg/kg orally for 21 days to establish a reproductive damage model.Then corresponding drugs were intragascasted for 30 days,and the weight change of mice was observed.The organ index,sperm survival rate of the testicles and epididymis,and oxidative damage in testicular was determined,,and the pathological structure of mice testicles was observed.Results The testis and epididymis indices in the mediumdose group showed a significant increase compared to the model group(P<005),the sperm motility in both the medium and highdose groups was significantly improved(P<005),and the superoxide dismutase(SOD)was significantly increased.The histological examination revealed a marked increase in the number of spermproducing cells in the testis of mice treated with Bletilla striata polysaccharides at medium and high doses.Conclusion Bletilla striata polysaccharides improved the testis and epididymis indices,sperm count,and motility in mice with ORNinduced reproductive damage,indicating its potential therapeutic effect in mitigating oxidative stressinduced reproductive system impairment.
