摘  要:目的:探讨总睾酮(TT)、雌二醇(E_(2))水平与玫瑰痤疮发生风险的潜在相关性。方法:采用两样本孟德尔随机化分析方法,通过全基因组关联研究数据库(GWAS)检索数据。将TT、E_(2)水平的相关遗传变异数据作为工具变量,玫瑰痤疮的相关遗传变异数据作为结局事件,采用逆方差加权法(IVW)、MR-Egger法、Simple Mode法、加权中位数法和Weighted Mode法等回归模型分别分析TT、E_(2)水平与玫瑰痤疮发生风险的相关性,并进行敏感性分析来验证结果的可靠性。结果:IVW结果显示,TT水平与发生玫瑰痤疮正相关(OR=2.217,95%CI:1.290~3.811,P<0.05),E_(2)水平与发生玫瑰痤疮无相关性(P>0.05)。ME-Egger结果显示,SNP不存在多效性(P>0.05),且不存在异质性及离群值,无单独的SNPs影响结果,结果具有稳健性。结论:TT水平升高与发生玫瑰痤疮有潜在相关性,E_(2)水平与发生玫瑰痤疮无相关性。Objective:To explore the potential correlation between the levels of total testosterone(TT),estradiol(E_(2))and the risk of rosacea.Methods:Two-sample Mendelian randomization analysis method was used for data retrieval using genome-wide association Study database(GWAS).The genetic variation data of TT and E_(2) levels were used as instrumental variables,and the genetic variation data of rosette acne were used as outcome events.Regression models such as inverse variance Weighted method(IVW),MR-Egger method,Simple Mode method,weighted median method and Weighted Mode method were used to analyze the correlation between the levels of TT,E_(2) and the risk of rosette acne,and sensitivity analysis was conducted to verify the reliability of the results.Results:IVW results showed that there was a positive correlation between TT level and the risk of rosacea(OR=2.217,95%CI:1.290~3.811,P<0.05),but not E_(2) level(P>0.05).The ME-Egger results indicated that SNPs did not have pleotropy(P>0.05),and there was no heterogeneity and outlier,and no single SNPs affected the results,so the results were robust.Conclusion:There is a potential correlation between increased TT and the risk of rosacea,and there is no statistical evidence that the risk of rosacea is related to the level of E_(2).
