摘 要:采用超临界CO_(2)技术制备栀子果油(ZGY),通过GC-MS分析其脂肪酸组成、CCK8法检测细胞活力、ELISA和qPCR检测细胞损伤下不同关键指标的变化。结果显示,ZGY含有44.85%亚油酸、36.26%油酸、15.17%棕榈酸,不饱和脂肪酸相对百分含量高达82.34%。与模型组相比,ZGY对LPS诱导RAW 264.7细胞产生NO的量有很强的抑制作用。经体积分数0.625%和1.25%ZGY同步处理下,可显著抑制UVB诱导的光损伤HaCaT细胞内炎症因子IL-6和IL-8 mRNA的表达量,提升屏障因子LOR、ZO-1、CLDN3 mRNA的表达量。因此,ZGY可减少NO的产生量,下调炎症因子mRNA的表达,上调屏障因子mRNA的表达,起到舒缓和修护的作用,为栀子果油的应用开发提供理论基础。The fatty acid composition of Gardenia jasminoides fruit oil(ZGY)with preparation by supercritical CO_(2) technology was analyzed through GC-MS.The cell viability was detected by CCK8 method,ELISA and qPCR were applied to detect different key indexes under cell injury.The results showed that the fatty acids of ZGY included 44.85%linoleic acid,36.26%oleic acid,15.17%palmitic acid and 82.34%unsaturated fatty acids.Compared with the model group,ZGY had a strong inhibitory on NO production in LPS-induced RAW 264.7 cells.After simultaneous treatment with 0.625%and 1.25%ZGY in volume fraction,it significantly inhibited the expression levels of IL-6 and IL-8 mRNA,and increased the expression levels of LOR,ZO-1 and CLDN3 mRNA in UVB irradiation-HaCaT cells.Therefore,ZGY can reduce the production of NO,down-regulate the expression of inflammatory factor mRNA,up-regulate the expression of barrier factor mRNA,and play a soothing and repairing role,which provides a theoretical basis for the application and development of ZGY.
