摘  要:癌细胞依靠糖酵解获得能量以供生长繁殖是癌症发展的核心环节。造成癌细胞有氧糖酵解增强的主要原因与糖酵解关键酶及其转运蛋白、信号通路、非编码RNA密切相关。近年来研究显示,多种中药单体(或其活性成分)及复方能够通过调控肿瘤细胞糖酵解发挥抗肿瘤作用,提升癌症中晚期患者的生存质量,延长生存期。本文以糖酵解为切入点,分别从通过靶向葡萄糖转运体,下调单羧酸转运蛋白、己糖激酶表达、磷酸果糖激酶的表达,调节丙酮酸激酶、乳酸脱氢酶的活性,抑制糖酵解过程;从myc、Ras等促癌基因及磷脂酰肌醇3-激酶/蛋白激酶B/哺乳动物雷帕霉素靶蛋白通路等代谢通路研究有氧糖酵解抗肿瘤机制;从微小RNA、长链非编码RNA等非编码RNA等三方面,总结近年来基于糖酵解的中医药抗恶性肿瘤机制,以期为中医药抗肿瘤研究提供参考。Cancer cells rely on glycolysis for energy to grow and reproduce,which is central to cancer development.The main reasons for the enhancement of aerobic glycolysis in cancer cells are closely related to key glycolysis enzymes,their transporters,signaling pathways and non-coding RNA.Recent studies have shown that a variety of traditional Chinese medicine(TCM)monomers(or their active ingredients)and compounds can exert anti-tumor effects by regulating the glycolysis of tumor cells,improve the quality of life and prolong the survival of patients with advanced cancer.By targeting glucose transporters,the expression of monocarboxylate transporter,hexokinase and phosphofructokinase is down-regulated,and the activities of pyruvate kinase and lactate dehydrogenase are regulated to inhibit the glycolysis process.From myc,Ras and promote cancer gene and phosphatidyl inositol 3 kinase/protein kinase B/mammals rapamycin target protein phosphatidylinositol3-kinase/Protein kinase B/mammaliantargetofrapamycin(PI3K/Akt/mTOR)pathway and other metabolic pathways to study the anti-tumor mechanism of aerobic glycolysis;from the perspective of microRNA(miRNA),longnoncoding RNA(lncRNA)and other non-coding RNAs,the anti-malignant tumor mechanism of TCM based on glycolysis in recent years was summarized,in order to provide references for the anti-tumor research of TCM.
