基于金属有机框架材料ZIF-8纳米酶酶解全蝎蛋白研究

摘  要:实验研究金属-有机框架材料水解纳米酶对动物药全蝎蛋白的酶解作用。首先从5种有机框架纳米材料中选出酶解效果最好的水解纳米酶,然后选用已知蛋白牛血清白蛋白(BSA)检测有机框架纳米材料是否具有蛋白酶样活性,并对其酶解工艺进行优化。通过比较5种有机框架纳米材料对全蝎蛋白的酶解效果,得出金属-有机框架纳米复合材料ZIF-8对全蝎蛋白的酶解效果最好。通过对比考察材料用量和酶解时间对酶解结果的影响,得出了最佳反应条件,6 mg的ZIF-8在60℃条件下反应15min的酶解效果最好。综上所述,ZIF-8作为纳米酶酶解过程快速简便,有望用于全蝎蛋白的水解。The enzymatic hydrolysis of animal drug scorpion protein by metal-organic framework material hydrolyzed nanozyme was studied.Firstly,the hydrolysis nanozyme with the best enzymatic hydrolysis effect was selected from five kinds of organic framework nanomaterials,and then the known protein bovine serum albumin(BSA)was used to detect whether the organic framework nanomaterials had protease-like activity,and the enzymatic hydrolysis process was optimized.By comparing the enzymatic hydrolysis effects of five organic framework nanomaterials on scorpion protein,it was concluded that ZIF-8 had the best enzymatic hydrolysis effect on scorpion protein.The optimum reaction conditions were obtained by comparing the effects of material dosage and enzymolysis time on the enzymolysis effects.The enzymolysis effect of 6 mg ZIF-8 at 60℃for 15 min was the best.In summary,ZIF-8 as a nanozyme has the fast and simple enzymatic hydrolysis process,and is expected to be used for scorpion protein hydrolysis.
