摘 要:利用超高效液相色谱-四级杆-静电场轨道阱高分辨质谱技术(UHPLC-Q-Orbitrap-HRMS)快速鉴定鼻渊舒口服液的主要化学成分,明确其药效物质基础。采用ACQUITY UPLCBEH C_(18)色谱柱(100 mm×2.1 mm,1.7μm),以流动相0.1%甲酸乙腈溶液(A)-0.1%甲酸水溶液(B)进行梯度洗脱,电喷雾离子源,HESI离子源,正负离子切换扫描模式。通过对照品、质谱数据库和参考文献比对,共鉴定出94种成分,包括42种黄酮类、13种萜类、7种酚酸类、6种香豆素类、8种生物碱类、4种有机酸类及14种其他成分。通过对典型化合物的质谱裂解规律分析,揭示其特征质谱裂解规律。该研究为鼻渊舒口服液的作用机制研究和质量控制提供了科学依据,并为其临床应用奠定了理论基础。The main chemical components of Bi-Yuan-Shu Oral Liquid were rapidly identified using ultra-high performance liquid chromatography coupled with quadrupole electrostatic field Orbitrap high-resolution mass spectrometry(UHPLC-Q-Orbitrap-HRMS),aiming to elucidate the material basis for its pharmacological effects.An ACQUITY UPLC BEH C_(18)column(100 mm×2.1 mm,1.7μm)was used,with gradient elution performed using 0.1%formic acid in acetonitrile(A)and 0.1%formic acid in water(B)as the mobile phases.Electrospray ionization(HESI)was applied with positive and negative ion switching mode.A total of 94 components were identified through comparison with reference standards,mass spectrometry databases and literature,including 42 flavonoids,13 terpenoids,7 phenolic acids,6 coumarins,8 alkaloids,4 organic acids and 14 other compounds.The characteristic fragmentation patterns of typical compounds were analyzed,revealing their specific fragmentation behaviors.The scientific evidence was provided for the investigation of the pharmacological mechanisms and quality control of Bi-Yuan-Shu Oral Liquid and a theoretical foundation for its clinical application was established.
