摘  要:目的:分析外源性神经生长因子激活Wnt3α/β-catenin信号通路促进骨折愈合的作用。方法:选取40只SPF级SD大鼠,10只大鼠作为对照组,30只构建胫骨骨折模型,共有27只大鼠建模成功,分为模型组9只、低剂量组9只、高剂量组9只。观察各组大鼠病理组织学、骨钙、骨磷水平、血液流变学指标、BMD、骨痂直径、血管生成相关因子、炎症因子水平、Wnt3α/β-catenin通路关键基因Wnt3a、β-catenin、GSK-3β的mRNA表达量、Wnt3α/β-catenin通路关键蛋白Wnt3a、β-catenin、GSK-3β的表达。结果:与对照组相比,另外三组骨钙、骨磷、BMD、血管生成相关因子、Wnt3α、β-catenin相关mRNA表达量、Wnt3α、β-catenin表达量降低,全血黏度、血浆黏度、炎症因子水平、GSK-3β相关mRNA表达量、GSK-3β表达量升高,与模型组相比,低、高剂量组骨钙、骨磷、BMD、骨痂直径、血管生成相关因子、Wnt3α、β-catenin相关mRNA表达量、Wnt3α、β-catenin表达量升高,全血黏度、血浆黏度、炎症因子水平、GSK-3β相关mRNA表达量、GSK-3β表达量降低;且高剂量组体现出最高骨钙、骨磷、BMD、骨痂直径、血管生成相关因子、Wnt3α、β-catenin相关mRNA表达量、Wnt3α、β-catenin表达量,最低全血黏度、血浆黏度、炎症因子水平、GSK-3β相关mRNA表达量、GSK-3β表达量(P<0.05)。结论:采用外源性神经生长因子对胫骨骨质大鼠干预,可促使大鼠骨折的愈合,其作用机制可能与激活Wnt3α/β-catenin信号通路有关,具有较好的使用效果。Objective:To analyze the effect of exogenous nerve growth factor on the activation of Wnt3α/β-catenin signaling pathway in promoting fracture healing.Methods:Select 40 SPF grade SD rats,10 rats as the control group,and 30 rats to construct a tibial fracture model.A total of 27 rats were successfully modeled and divided into a model group of 9 rats,a low-dose group of 9 rats,and a high-dose group of 9 rats.Observe the pathological histology,bone calcium and phosphorus levels,hemorheological indicators,BMD,callus diameter,angiogenesis related factors,inflammatory factor levels,mRNA expression levels of key genes Wn3a,β-catenin,and GSK-3βin the Wnt3α/β-catenin pathway,and expression levels of key proteins Wnt3a,β-catenin,and GSK-3βin the Wnt3α/β-catenin pathway in each group of rats.Results:Compared with the control group,the other three groups showed a decrease in bone calcium,bone phosphorus,BMD,angiogenesis related factors,Wnt3α,β-catenin related mRNA expression,Wnt3α,β-catenin expression,and an increase in whole blood viscosity,plasma viscosity,inflammatory factor levels,GSK-3βrelated mRNA expression,and GSK-3βexpression.Compared with the model group,the low and high dose groups showed an increase in bone calcium,bone phosphorus,BMD,callus diameter,angiogenesis related factors,Wnt3α,β-catenin related mRNA expression,Wnt3α,β-catenin expression,and a decrease in whole blood viscosity,plasma viscosity,inflammatory factor levels,GSK-3βrelated mRNA expression,and GSK-3βexpression;And the high-dose group showed the highest levels of bone calcium,bone phosphorus,BMD,callus diameter,angiogenesis related factors,Wnt3α,β-catenin related mRNA expression,Wnt3α,β-catenin expression,and the lowest levels of whole blood viscosity,plasma viscosity,inflammatory factor levels,GSK-3β-related mRNA expression,and GSK-3βexpression(P<0.05).Conclusion:The intervention of exogenous nerve growth factor on tibial bone can promote the healing of fracture in rats,the mechanism of which may be related to the activ
