黄海西南部海门潮间带有孔虫Ammonia分子系统学研究初探

摘  要:提要研究对采自于黄海西南部海门潮间带表层沉积物中的底栖有孔虫进行了分子生物学研究尝试。通过对底栖有孔虫的实验培养与形态观察,对群落中含量最高的Ammonia spp.,根据其壳体形态特征挑选六个典型类型、21个个体进行了分析。通过DNA提取、PCR扩增、单克隆培养与测序,获得了这些有孔虫个体大量的核糖体小亚基(Ribosomal Small Subunit,SSU) rDNA序列。本文报道了HM03个体的19个克隆DNA序列、HM21个体的6个克隆DNA序列,以及HM02和HM04-HM20个体各自一个克隆DNA序列。分子系统发育分析结果表明,除了HM21个体与其他个体亲缘关系较远外,其他不同形态特征的分子与潮间带低盐分子Ammonia catesbyana(d'Orbigny,1839),Ammonia aomoriensis (Asano,1951),Ammonia confertitesta Zheng,1978和Ammonia sp.T6(Hayward et al.,2004)等亲缘关系紧密,形成一独立系群(Clade A),推测其为同种异名。因为A.confertitesta是邻近地区研究最早报道建立的种名,因此本潮间带常见Ammonia分子暂时使用A.confertitesta种名。观察显示,A.confertitesta不同个体的末圈房室数量、脐部及缝合线等特征可以有较大程度的变化。另外,HM21个体根据其壳体形态特征初步鉴定为Ammonia convexidorsa Zheng,1978,系统树支持其SSU rDNA序列为一独立系群(Clade B),该系群SSU rDNA序列为首次报道。本研究结合已有的Ammonia多个相关属种系统演化关系,对浅海近岸环境中最常见分子分类进行了厘定、并提出使用建议,为微体古生物学与古环境应用提供重要基础。Living benthic foraminifers from the Haimen intertidal zone in the southwestern Yellow Sea were sampled to investigate the classification of Ammonia spp.based on the DNA phylogenetic analysis.Through the laboratory culture and morphological observation,the Ammonia specimens were subdivided into 6 distinct groups.Additionally,21 specimens(HM01-HM21)of them were selected for the SSU rDNA sequence analyses.Following DNA extraction,PCR amplification,cloning and sequencing,a large amount of DNA sequences were obtained,with the exception of HM01.In total,we report 19 clone sequences from HM03,six from HM21 and one sequence each from HM02 and HM04-HM20.These sequences were conducted phylogenetic analysis together with other 16 related Ammonia sequences from the GenBank.The phylogenetic tree suggests that HM02-HM20 form one distinct group(Clade A),together with Ammonia catesbyana(d'Orbigny,1839),Ammonia aomoriensis(Asano,1951),Ammonia confertitesta Zheng,1978 and Ammonia sp.T6(Hayward et al.,2004).In contrast,the HM21 clones represent another separate group(Clade B).Due to its first report and nomination in the neighbor research region(Zheng et al.,1978),A.confertitesta is tentatively adopted for the most common Ammonia species in the Haimen intertidal zone.Based on our findings,A.confertitesta exhibits a wide range of morphological variation in the chamber number,umbilical opening,folia shape and sutural furrows. Furthermore, the positions of different clone sequences from HM03 in the phylogenetic tree suggestthat the A. confertitesta, A. catesbyana and A. aomoriensis be synonymous. Based on the morphology, the specimen ofHM21 is assigned to Ammonia convexidorsa Zheng, 1978, with its SSU rDNA sequences first reported here. However,more samples need to be collected for its morphologic and DNA phylogenetic analyses.The molecular phylogenetic study on the Ammonia spp. provides important insights on the intraspecificmorphological variation and DNA sequence diversity which will benefit future foraminiferal taxonomy and paleo
