摘 要:目的基于孟德尔随机化(MR)探讨肠道菌群与代谢综合征(MS)的因果关系。方法从英国基因史研究的全基因组相关研究数据库中提取与肠道菌群和MS相关的单核苷酸多态性(SNP)位点作为干预基因,以工具变量SNP为索引,构建工具变量-暴露因素-结局变量三者的关系。采用MR-Egger、加权中位数、逆方差加权、简单模式和加权模式5种MR分析方法,探究肠道菌群与MS的因果关系。结果逆方差加权结果显示,拟杆菌目-S24-7、双歧杆菌科、粪球菌属1、双歧杆菌目和放线菌门是MS的保护因素(OR=0.936,95%CI 0.882~0.992,P=0.026;OR=0.928,95%CI 0.868~0.992,P=0.028;OR=0.922,95%CI 0.860~0.988,P=0.021;OR=0.928,95%CI 0.868~0.992,P=0.028;OR=0.935,95%CI 0.878~0.996,P=0.037),韦荣球菌科、奥尔森菌属和厚壁菌门是MS的危险因素(OR=1.055,95%CI 1.004~1.108,P=0.034;OR=1.046,95%CI 1.009~1.085,P=0.015;OR=1.063,95%CI 1.006~1.124,P=0.031)。另外4种方法与逆方差加权的方向一致。结论厚壁菌门、放线菌门、双歧杆菌目、拟杆菌目-S24-7、双歧杆菌科、韦荣球菌科、粪球菌属1和奥尔森菌属与MS之间存在因果关联。Objective To explore the causal correlation between gut microbiota and metabolic syndrome(MS)based on Mendelian randomization(MR).Methods The single nucleotide polymorphisms(SNP)loci related to gut microbiota and MS were extracted from the genome wide related study database of genetic history research in the UK as intervention genes.The instrumental variable SNP was used as an index to construct the relationship between instrumental variable-exposure factor-outcome variable.Five MR analysis methods,including MR-Egger,weighted median,inverse variance weighted,simple mode and weighted mode,are used to explore the causal relationship between gut microbiota and MS.Results The inverse variance weighted analysis results showed that Bacteroidales S24-7 group,Bifidobacteriaceae,Coprococcus 1,Bifidobacteriales and Actinobacteria were protective factors for MS(OR=0.936,95%CI 0.882-0.992,P=0.026;OR=0.928,95%CI 0.868-0.992,P=0.028;OR=0.922,95%CI 0.860-0.988,P=0.021;OR=0.928,95%CI 0.868-0.992,P=0.028;OR=0.935,95%CI 0.878-0.996,P=0.037),Veillonellaceae,Olsenella and Firmicutes were risk factors for MS(OR=1.055,95%CI 1.004-1.108,P=0.034;OR=1.046,95%CI 1.009-1.085,P=0.015;OR=1.063,95%CI 1.006-1.124,P=0.031).The other four methods agreed with the direction of the inverse variance weighting.Conclusion There are causal associations of gut microbiota Firmicutes,Actinobacteria,Bifidobacteriales,Bacteroidales S24-7 group,Bifidobacteriaceae,Veillonellaceae,Coprococcus 1 and Olsenella with MS.
