摘 要:为了解广东地区鹅源新城疫病毒(NDV)遗传进化及致病性情况,于2019年从广东肇庆、佛山地区疑似NDV感染鹅的组织病料中进行鹅源NDV分离鉴定并选取一株MDT最小的毒株进行病毒对雏鹅的致病性试验。结果显示:共分离6株Ⅻb亚型鹅源NDV毒株,6株毒株间核苷酸和氨基酸相似性最高,但与基因Ⅶ型、La Sota等常用疫苗株相似性较低。F蛋白和HN蛋白部分功能位点氨基酸残基出现不同程度的变异。致病性试验结果表明基因Ⅻb亚型Goose/GD/F424/2019毒株对雏鹅有较强的致病性,死亡率高达87.5%,并出现脾脏肿大,呈“树枝状”样充血、腺胃乳头出血、十二指肠黏膜出血、胰脏变性出血、脑组织充血及水肿等典型病理变化症状。研究表明:从广东地区分离的6株鹅源NDV均是Ⅻb亚型且对雏鹅有较强的致病性,为进一步研究广东地区鹅源NDV流行、进化及相关疾病的防控奠定基础。To investigate the genetic evolution and pathogenicity of Newcastle Disease Virus(NDV)from geese in the Guangdong region,an experiment in 2019 involved the isolation and identification of goose-derived NDV from tissue specimens of suspected NDV-infected geese in Zhaoqing and Foshan areas.A strain with the smallest mean death time(MDT)was selected for pathogenicity testing in goslings.The results showed that six subtypeⅫb NDV strains from geese were isolated.These six strains had the highest nucleotide and amino acid sequence similarity between them,but showed lower similarity with genotypeⅦand commonly used vaccine strains like La Sota.Variations of different degrees were observed at some functional sites of the F protein and HN protein amino acid residues.Pathogenicity tests indicated that the subtypeⅫb strain Goose/GD/F424/2019 exhibited strong pathogenicity in goslings,with a high mortality rate of 87.5%,and displayed typical pathological changes such as splenomegaly with“branch-like”congestion,bleeding of proventricular spicules,bleeding of the duodenal mucosa,pancreatic degeneration and hemorrhage,and congestion and edema of brain tissue.The study indicated that all six isolated NDV strains from geese in the Guangdong region belonged to theⅫb subtype and were highly pathogenic to goslings,laying a foundation for further research on the prevalence,evolution,and disease control of goose-derived NDV in the Guangdong area.
