摘  要:以伏牛山野生黄精的种子为试验材料,建立其组培快繁体系并对组培苗质量进行评估。结果表明,伏牛山野生黄精种子萌发最佳培养基为1/2 MS+0.5 mg/L ABA,丛生芽诱导最佳培养基为MS+4.0 mg/L 6-BA+0.2 mg/L NAA+0.5 mg/L GA3,丛生芽增殖最佳培养基为MS+3 mg/L 6-BA+0.1 mg/L TDZ+0.5 mg/L 2,4-D+0.4 mg/L NAA,生根最佳培养基为1/2 MS+0.5 mg/L NAA+0.2 mg/L IBA;组培苗分级标准为Ⅰ级苗:株高≥4.2 cm、叶片≥5个,Ⅱ级苗:3 cm<株高<4.2 cm、2<叶片数<5,Ⅲ级苗株高≤3 cm、叶片数≤2个,移栽后成活率为88%。The tissue culture rapid propagation system was established and the quality of the tissue culture seedlings was evaluated with the wild Polygonatum sibiricum seeds from Funiu Mountain as the research material.The results showed that the best medium for seed germination of Polygonatum sibiricum from Funiu Mountain was 1/2 MS+0.5 mg/L ABA,the best medium for cluster bud induction was MS+4.0 mg/L 6-BA+0.2 mg/L NAA+0.5 mg/L GA 3,and the best medium for cluster bud proliferation was MS+3 mg/L 6-BA+0.1 mg/L TDZ+0.5 mg/L 2,4-D+0.4 mg/L NAA,the best rooting medium was 1/2 MS+0.5 mg/L NAA+0.2 mg/L IBA.The classification standard of tissue culture seedlings was GradeⅠ:plant height≥4.2 cm,leaves≥5,GradeⅡ:3 cm<plant height<4.2 cm,2<number of leaves<5,GradeⅢ:plant height≤3 cm,number of leaves≤2,the survival rate after transplantation was 88%.
